扶正康复合剂与顺铂联用抑制移植性肺癌的实验研究 被引量:4

摘要 目的:研究扶正康复合剂对移植性肺癌的抑制作用,并探讨其作用机制。方法:将48只NOD/SCID裸鼠随机分为8组:模型对照组,顺铂组,扶正康复合剂低、中、高剂量组,顺铂+扶正康复合剂低、中、高剂量组,共8组,每组6只,皮下接种H1975细胞建立移植性裸鼠肺癌模型,接种15 d后开始给药,连续给药7 d,称取体重及瘤重,计算抑瘤率,ELISA法检测裸鼠血清BUN、Cr、TNF-α、IFN-γ、IL-2、IL-6的表达水平,免疫组化法检测瘤体组织VEGF、Bcl-2、Bax、Survivin的表达情况,q-PCR法检测瘤体组织VEGF、Bcl-2、Bax、Survivin mRNA的表达水平。结果:扶正康复合剂高剂量组、顺铂组、顺铂+扶正康复合剂低、中、高剂量组与模型对照组相比:荷瘤裸鼠瘤质量显著减轻(P<0.05),血清TNF-α水平显著升高(P<0.05),IL-2水平显著升高(P<0.05),血清IL-6水平显著降低(P<0.05),Bcl-2表达量显著减少,Bax表达量增加。其中顺铂+扶正康复合剂高剂量给药组裸鼠抑瘤率最高,两者具有协同作用。与顺铂组相比:顺铂+扶正康复合剂高剂量组裸鼠血清Cr、BUN水平显著降低(P<0.01),顺铂+扶正康复合剂中、高剂量组裸鼠血清TNF-α、INF-γ水平显著升高(P<0.01);顺铂+扶正康复合剂高剂量组Bcl-2、Survivin、VEGF表达量降低,Bax表达量增加。结论:扶正康复合剂与顺铂联用能够降低顺铂的肾损伤作用,调节机体的Th1/Th2细胞免疫平衡,通过抑制Bcl-2、Survivin、VEGF并促进Bax的表达,进而促进肺癌细胞的凋亡。 Objective : To study the anti - lung cancer effect of Fuzheng Kangfu Mixture and investigate its possible mechanisms. Methods : Forty - eight NOD/SCID mice were randomly divided into 8 groups : model control group, cispla- tin group, Fuzheng Kangfu Mixture low, middle and high doses groups, cisplatin + Fuzheng Kangfu Mixture low, middle and high dose groups. H1975 cells were given subcutaneous inoculation to establish nude - mouse transplanted lung tumor model. After 15 days, continuous administration drugs for 7 days, weigh body and tumor weights and calculate the tumor inhibition rate. ELISA method was used to detect the serum BUN, Cr, TNF - α, IFN - γ, IL - 2 and IL - 6 expres- sions. The immunohistochemistry method was used to detect the expressions of VEGF, Bcl - 2, Bax, and survivin in tumor. The q -PCR method was used to detect the VEGF, Bcl -2, Bax and survivin mRNA expression levels in tumor tissue. Results: Compared with model group, Fuzheng Kangfu Mixture high doses, cisplatin and cisplatin + Fuzheng Kangfu Mixture low, middle and high doses could inhibit the growth of lung tumor( P 〈 0.05 ) , increase the serum levels of TNF - α and IL - 2 ( P 〈 0. 05 ), decrease the serum level of IL - 6 ( P 〈 0.05 ) and decrease the expressions of Bcl - 2 while increase the expression of Bax. Among them, the tumor inhibition rate of cisplatin + Fuzheng Kangfu Mixture high doses group was the highest and both had synergistic effect. Compared with cisplatin group, the serum levels of Cr and BUN were significantly decreased in cisplatin + Fuzheng Kangfu Mixture high doses group( P 〈 0.01 ). Fuzheng Kangfu Mixture middle and high doses combined with cisplatin could promote the expressions of TNF - α and IFN - γ ( P 〈 0.01 ). Compared with cisplatin group, the expressions of Bcl -2, survivin and VEGF in cisplatin + Fuzheng Kangfu Mixture high doses group were significantly increased while the expressions of Bax were significantly decreased.
