摘要
目的:观察葛根素对妊娠期糖尿病模型大鼠脑组织氧化应激和细胞凋亡的影响。方法:糖尿病模型大鼠随机分为妊娠组、模型组、二甲双胍组和葛根素组,每组18只;另取18只正常大鼠作为正常组。二甲双胍组大鼠给予二甲双胍(200 mg·kg^(-1))灌胃;葛根素组大鼠给予葛根素(160 mg·kg^(-1))灌胃,正常组、妊娠组和模型组均给予等体积的生理盐水灌胃,连续14 d。检测各组大鼠不同时间空腹血糖、血清总胆固醇(total cholesterol,TC)、三酰甘油(triglyceride,TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterin,LDL-C)、高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)、总抗氧化能力(total antioxidant capacity,T-AOC)、丙二醛(malondialdehyde,MDA)的水平,脑组织中超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、过氧化氢酶(catalase,CAT)的水平以及大鼠脑细胞凋亡情况。结果:(1)二甲双胍组和葛根素组大鼠空腹血糖水平明显低于模型组,且血糖水平呈持续性下降,差异有统计学意义(P<0.05);(2)二甲双胍组和葛根素组大鼠TC、TG、LDL-C和MDA水平明显低于模型组,HDL-C和T-AOC水平高于模型组,且葛根素组大鼠TC、TG、LDL-C和MDA水平低于二甲双胍组,HDL-C和T-AOC水平高于二甲双胍组,差异均有统计学意义(P<0.05);(3)二甲双胍组和葛根素组大鼠脑组织中SOD、GSH-Px、CAT水平均明显高于模型组,且葛根素组明显高于二甲双胍组,差异均有统计学意义(P<0.05);(4)葛根素组大鼠脑细胞凋亡率明显低于模型组和二甲双胍组,差异有统计学意义(P<0.05)。结论:葛根素可以降低妊娠期糖尿病大鼠血糖和血脂,并且能够明显提高大鼠抗氧化能力,可通过提高SOD、GSH-Px和CAT水平来减少脑组织的损伤,预防脑损伤。
Objective: To study the effect of Puerarin on oxidative stress and apoptosis in brain tissue of gestational diabetes rats.Methods: Rats with Diabetes were randomly divided into normal group,pregnancy group,model group,metformin group and puerarin group,with 18 rats in each group. The rats in metformin group were given intragastric administration of metformin( 200 mg·kg-1) and the rats in puerarin group rats were given intragastric administration of Puerarin( 160 mg· kg-1). Rats in normal group,pregnancy group and the model group were given equal volume saline,for 14 consecutive days. Fasting blood glucose,total cholesterol( TC),triglyceride( TG),low density lipoprotein cholesterin( LDL-C),high density lipoprotein cholesterol( HDL-C),total antioxidant capacity( T-AOC),malondialdehyde( MDA) levels,superoxide dismutase( SOD) in brain tissue,Levels of glutathione peroxidase( GSH-Px),catalase( CAT),and apoptosis of rat brain cells were measured at different times in each group.Results:① The fasting blood glucose levels in the metformin group and the puerarin group were significantly lower than those in the model group,and the blood glucose level was continuously decreased,with the difference being statistically significant( P〈0. 05).② The levels of TC,TG,LDL-C and MDA The metformin group and the puerarin group were significantly lower than those of the model group. The levels of HDL-C and T-AOC were higher than those of the model group. The levels of TC,TG,LDL-C and MDA in the puerarin group were lower than those in the metformin group,while the levels of HDL-C and T-AOC were significantly higher than those of metformin group( P〈0. 05). The levels of SOD,GSH-Px and CAT in brain tissue of 3 metformin group and puerarin group were significantly higher than those in model group and that in the puerarin group was significantly higher than that in the metformin group,with the difference being statistically significant( P〈0. 05).
