葛根素对妊娠期糖尿病大鼠氧化应激损伤保护作用的研究 被引量:9

Effects of puerarin against oxidative stress injury in gestational diabetic rats

摘要 目的:研究葛根素(Puerarin)对妊娠期糖尿病大鼠氧化应激损伤的保护作用。方法:将80只通过腹腔注射链脲佐菌素制备的妊娠5天的糖尿病大鼠随机分为4组:妊娠期糖尿病模型对照组、葛根素(40、80和160 mg/kg)治疗组和盐酸二甲双胍(200 mg/kg)阳性药对照组,每组16只大鼠,另取16只同期妊娠5天的大鼠作为正常妊娠对照组以及16只同龄非妊娠大鼠作为正常非妊娠对照组。每天经灌胃给药1次,妊娠期糖尿病大鼠、正常妊娠对照组和正常非妊娠对照组分别给予等体积的生理盐水,疗程为2周。分别于给药前、给药治疗第7天和第14天测定空腹血糖水平;给药治疗2周后,分别检测各组大鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)和碱性磷酸酶(ALP)活性,丙二醛(MDA)含量以及总抗氧化能力(T-AOC)水平;测定肝脏组织匀浆中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的活性以及丙二醛(MDA)的含量;通过苏木精-尹红(HE)染色观察肝脏组织病理形态学改变状况。结果:与正常非妊娠糖尿病大鼠相比,正常妊娠糖尿病大鼠血糖水平,血清中ALT、AST、ALP活性,MDA含量以及TAOC水平均无显著性变化;肝组织匀浆中SOD、CAT活性和MDA含量均无显著性差异;肝组织均未出现病理形态学改变。与正常妊娠糖尿病大鼠相比,妊娠期糖尿病模型对照组大鼠血糖显著升高,血清中ALT、AST、ALP活性和MDA含量显著升高,T-AOC水平显著降低;肝组织匀浆中SOD、CAT活性显著降低且MDA含量显著升高;肝组织出现明显的病理形态学改变。与妊娠期糖尿病模型对照组相比,葛根素(80、160 mg/kg)治疗组大鼠血清中ALT、AST活性和MDA含量均显著降低,肝脏组织匀浆中SOD且MDA含量显著降低;葛根素160 mg/kg治疗组大鼠血糖水平显著降低,血清中ALP活性显著降低、T-AOC水平显著升高,肝脏组织中CAT活性显著升高;葛根素160 mg/kg治疗组肝脏组织病理学改变明显改� Objective: To investigate the effects of puerarin against oxidative stress injury in gestational diabetic rats. Methods: 64 gestational diabetic rats made by intraperitonal injecting STZ( 35 mg / kg) were randomly devided into five groups: gestational diabetic model group,puerarin( 40,80,and 160 mg / kg) treated groups,which was 16 expremental animals every group. Selected 20 rats with gestation for 5 days as normal gestation group and 20 rats with non-gestation as normal non-gestation group. The drugs were given by intragastric administration for two weeks,once a day. At 0,7,14 days after experiment,the level of plasma glucose of every group was determined. Two weeks later,the activity of ALT,AST,ALP in serum were determined; the content of MDA and the level of T-AOC in serum was determined; the activity of SOD,CAT and the content of MDA in hepatic tissue were also determined; and the histopathological changes of hepatic tissue were observed by HE staining. Results: Compared with the normal non-gestation group,the level of plasma glucose of the normal gestation group was not significantly different( P〈0. 05); the activity of ALT,AST,ALP,the content of MDA and the level of T-AOC in serum,and the activity of SOD,CAT and the content of MDA in hepatic tissue were also not significantly different( P〈0. 05). Compared with the normal gestation group,the plasma glucose level,the activity of ALT,AST,ALP and the content of MDA in serum of the gestational diabetic model group were significantly increased( P〈0. 01); the activity of SOD,CAT and the content of MDA in hepatic tissue were also significantly diffierent( P〈0. 01). Compared with the gestational diabetic model group,the plasma glucose level of puerarin 160 mg/kg treatment group was significantly decreased( P〈0. 01); the activity of ALT,AST and the content of MDA of puerarin( 80 and 160 mg / kg) treatment groups were significantly decreased( P〈0. 05,P〈0. 01),the activity of SOD in hepatic tissue were signific
