人羊膜上皮干细胞来源的外泌体促进小鼠神经干细胞向神经元方向分化 被引量:2

Human Amniotic Epithelial Stem Cells-Derived Exosomes Promote Neuronal Differentiation of Mouse Neural Stem Cells

摘要 人羊膜上皮干细胞(human amniotic epithelial stem cells,h AECs)能促进损伤神经元的修复和再生,但是否影响神经干细胞(neural stem cells,NSCs)的分化却很少报道。该研究首次发现h AECs能促进小鼠NSCs(mouse NSCs,m NSCs)向神经元方向分化。干细胞来源的外泌体(Exos)仍保留着干细胞的许多特性,因此,该研究探讨了h AECs来源的外泌体(h AECs-Exos)对m NSCs分化的影响。首先,采用超速离心的方法得到了纯度较高的h AECs-Exos,然后将不同浓度的h AECs-Exos与m NSCs共培养。结果发现,与没有h AECs-Exos的对照组相比,200 ng/m L h AECs-Exos能明显促进m NSCs向成熟神经元分化,主要表现在Neu N阳性细胞所占比例明显增高。研究者推测,h AECsExos保留了h AECs的一些特性,其含有的神经营养因子、生长因子、miRNAs等活性物质可能参与了微环境的调控,从而促进m NSCs向神经元方向分化。因此,h AECs-Exos可能促进内源或外源NSCs的神经分化,从而促进损伤或退化神经元的再生,这也为将来h AECs-Exos的临床应用提供研究基础。 Human amniotic epithelial stem cells(h AECs) have been reported to promote the regeneration of damaged neurons and the restoration of neurologic function, but whether h AECs can promote the neuronal differentiation of neural stem cells(NSCs) is rarely mentioned. In this present study, they found that h AECs could promote the neuronal differentiation of mouse NSCs(m NSCs). Additionally, stem cells-derived exosomes(Exos) can remain some characteristics of stem cells. They determined to investigate the effect of h AECs-derived exosomes(h AECs-Exos) on the neuronal differentiation of m NSCs. Firstly, h AECs-Exos were extracted byultracentrifugation. Then, h AECs-Exos at various concentrate were cocultured with m NSCs. The results suggested that the group with 200 ng/m L h AECs-Exos could significantly promote the neuronal differentiation of m NSCs compared with the control group with no h AECs-Exos, as demonstrated by the higher percentages of Neu N positive neurons derived from m NSCs. We hypothesized that the h AECs-Exos might retain some characteristics of h AECs. It provided a suitable microenvironment for the neuronal differentiation of m NSCs attributing to neurotrophic factors, growth factors, miRNAs and other active components. This could facilitate the neuronal differentiation of endogenous or exogenous NSCs, and thus contributed to the regeneration of damaged or degenerated neurons. This study will provide a preclinical study for the h AECs-Exos in regenerative medicine.
