实时荧光单引物等温扩增技术检测牛乳中的金黄色葡萄球菌 被引量:1

Detection of Staphylococcus aureus using a real-time single primer isothermal amplification in milk

摘要 研究建立了一种实时荧光单引物等温扩增(RF-SPIA)技术以检测牛乳中的金黄色葡萄球菌。针对金黄色葡萄球菌的耐热核酸酶(nuc)基因设计引物,对金黄色葡萄球菌进行特异性检测。结果表明,6株金黄色葡萄球菌的检测结果为阳性,而其他17株非金黄色葡萄球菌的检测结果均为阴性。RF-SPIA检测金黄色葡萄球菌纯培养物的灵敏度为2.0×101 CFU/m L,检测人工污染牛乳的检出限为6.0×101 CFU/m L。该方法特异性强、灵敏度高,能够实现对金黄色葡萄球菌的实时、快速检测,为金黄色葡萄球菌的检测提供了一条新途径。 The objective of this study was to establish a real-time fluorescence single primer isothermal amplification (RF-SPIA) for the detection of Staphylococcus aureus in milk. The SPIA primer was designed for specific detection of Staphylococcus aureus based on thermonuclease (nuc) gene. 6 strains of Staphylococcus aureus were shown to be positive using the RF-SPIA assay, whereas, 17 strains of non-Staphylococcus aureus in this study were negative. The sensitivity and the detection limit of RF- SPIA assay were 2.0×10^1 CFU/mL in pure culture and 6.0×10^1 CFU/mL in milk, respectively. The method provided strong specificity and sensitivity for the real-time rapid detection of Staphylococcus aureus, which could serve as a new wav for the detection of Staohvlococcus aureus
