摘要
利用化学合成对高效氯氟氰菊酯分子结构进行改造,制备了相应的半抗原,并与载体蛋白偶联合成人工抗原,免疫动物,制备单克隆抗体。在筛选高效氯氟氰菊酯单克隆抗体的基础上,建立酶联免疫检测方法,并研制出检测苹果、大豆、蔬菜中高效氯氟氰菊酯残留的试剂盒,蔬菜选择了莲花白作为检测对象。该试剂盒的灵敏度为1μg/L,对苹果、大豆、莲花白样本的最低检测限分别可达到20、30、60μg/kg;添加回收率68.8%~105.3%,批内变异系数为7.1%~13.2%;批间变异系数为7.9%~13.1%。对实际样本的检测结果与气相色谱法基本一致。该方法快速、灵敏、可靠,为水果、蔬菜中高效氯氟氰菊酯的残留检测提供了便捷、准确的分析检测手段。
The molecular structure of cyhalothrin was modified by chemical synthesis, and the corresponding hapten was prepared, and the artificial antigen was synthesized by coupling with the carrier protein, and the animal was immunized and the monoclonal antibody was prepared. Based on the screening of high performance cyhalothrin monoclonal antibody, an enzyme linked immunoassay was established, and a reagent kit for the detection of high performance cyhalothrin residues in apples, corn and vegetables was developed. The sensitivity of the kit is 1 g/L. The minimum detection limit for apple, corn and vegetable samples can reach 20, 30, and 60 μg/kg, respectively, and the coefficient of variation of the coefficient of variation is 7.1% ~ 13.2%, and the coefficient of variation is 7.9% ~ 13.1%. The results of actual samples are basically the same as those of gas chromatography. The method is rapid, sensitive and reliable, and provides a convenient and accurate analytical method for residue detection of highly effective cyhalothrin in fruits, vegetables and grains.
