微小RNA-21在肝癌组织的表达及其临床意义 被引量:8

Expression and clinical significance of microRNA -21 in hepatocellular carcinoma tissue samples

摘要 目的探讨微小RNA(miR)-21在肝癌组织中的表达及其临床意义。方法选择81例肝癌组织及癌旁组织和20例正常肝组织标本。实时定量反转录聚合酶链反应(RT.qPCR)法检测miR-21表达,分析miR-21表达与患者临床病理特征及预后的关系。结果与正常肝组织比较,miR-21肝癌组织中表达升高了(6.34±0.23)倍(P=0.003),在癌旁组织中升高(3.444-0.12)倍(P=0.020)。miR-21表达与肿瘤直径(X^2=11.034,P=0,001)、转移(X^2=12.081,P=0.001)、血管侵犯(X^2=13.372,P=0.000)及美国癌症联合委员会(AJCC)分期(X^2=12.175,P=0.000)明显相关,而与性别(X^2=0.290,P=0.590)、年龄(X^2=0.036,P=0.850)、肿瘤位置(X^2=0.593,P=0.441)、肿瘤分化(X^2=2.729,P=0.099)、肿瘤数量(X^2=1.597,P=0.206)及甲胎蛋白(AFP,X^2=0.279,P=0.597)无明显相关。肝癌组织miR-21高表达的患者5年生存率明显低于低表达患者(27.2%比10.7%,P=0.003),miR-21高表达是影响肝癌患者总体生存的独立风险因素(P=0.001)。结论miR-21在肝癌组织中表达明显上调,并且与肿瘤转移明显相关,参与了肝癌的发展过程。 Objective To detect the expression of microRNA (miRNA, miR) -21 in hepatocel- lular carcinoma (HCC) tissue samples and to explore its association with clinicopathologic features. Methods The real - time quantitative reverse transcriptase - polymerase chain reaction ( RT - qPCR ) analysis was used to detect the expression of miR - 21 in tissue specimens from HCC and the adjacent non- cancerous hepatic tissues collected from 81 patients who underwent surgical treatment for primat3, HCC and in normal liver tissues collected from 20 patients. Statistically, the association between the ex- pression of miR - 21 and clinicpathologic characteristics and prognosis of patients was analyzed. Results The expression of miR - 21 was up - regulated in HCC [ (6. 34 ± 0. 23 ) fold, P = 0. 003 ] and the adjacent non -cancerous hepatic tissues [ (3.44 ±0. 12) fold, P =0. 0201 as compared with that in the normal liv- er tissues. The results showed that high expression of miR -21 was strongly correlated with the characteris- tics of tumor size (X2 = 11. 034, P = 0. 001 ), metastasis (X^2= 12. 081, P = 0. 001 ), venous invasion (X^2 = 13. 372, P = 0. 000) and American Joint Committee on cancer (AJCC) TNM stage (X^2 = 12. 175, P = 0. 000). There were no significant associations between miR -21 expression and gender (Xz = 0. 290, P =0. 590), age (X^2 =0. 036, P =0. 850), tumor location(X^2 =0. 593, P =0. 441), tumor grade (X^2 = 2. 729, P = 0. 099), tumor number (X^2 = 1. 597, P = 0. 206) and alpha fetal protein ( AFP, X^2 = 0. 279, P = 0. 597 ). Patients with high miR -21 expression had significantly poorer overall survival (27.2% vs. 10. 7%, P =0. 003). High miR -21 expression was independent prognostic factors for overall survival (P = 0. 001 ). Conclusion The expression of miR - 21 is highly up - regulated in HCC tissues, which is correlated significantly with the process of metastasis and unfavorable prognosis.
