摘要
目的:研究肝细胞肝癌(HCC)大鼠癌细胞微小RNA-21(microRNA-21)含量与Toll样受体2(TLR2)、TLR4、核因子κB(NF-κB)水平的相关性.方法:40只SD大鼠随机分为对照组、模型组,各20只,模型组腹腔注射二乙基亚硝胺建立肝癌模型,对照组腹腔注射等量生理盐水,3次/周,连续12周.于12周末处死大鼠,解剖摘取肝脏,观察肝脏形态,行HE染色,采用RT-PCR检测microRNA-21、TLR2、TLR4、NF-κB mRNA表达,采用pearson相关性分析microRNA-21含量与TLR2、TLR4、NF-κB水平的相关性.结果:肝癌大鼠呈病态特征,精神状态、饮食活动、毛发生长、体重等异常,死亡大鼠较多.HE染色结果显示,肝癌大鼠肝组织呈水肿状态,色泽异常,表面粗糙,可见散在瘀斑,边缘变钝,质地翠硬易破裂,光镜可见肝小叶结构紊乱,肝小叶内肝细胞萎缩,大量坏死液化,中性粒细胞浸润.RT-PCR结果显示,模型组microRNA-21含量明显高于对照组,有统计学差异;模型组TLR2、TLR4、NF-κB mRNA表达水平明显高于对照组,有统计学差异;肝癌细胞中microRNA-21含量与TLR2、TLR4、NF-κB水平均呈正相关.结论:microRNA-21、TLR2、TLR4、NF-κB在肝癌大鼠癌细胞中表达上调,且microRNA-21含量与TLR2、TLR4、NF-κB水平呈正相关.
Objective To investigate the correlation between microRNA-21(mRNA-21)content and the levels of Toll-like receptor 2(TLR2),TLR4 and nuclear factor-kappa B(NF-κB)in hepatocellular carcinoma(HCC)rats.Methods 40 SD rats were randomly divided into control group and model group,20 each.The model group was intraperitoneally injected with di-ethylnitrosamine to establish HCC model.The control group was intraperitoneally injected with the same amount of normal saline 3 times a week for 12 weeks.At the end of the experiment,the liver morphology was observed.HE staining was performed.The expressions of microRNA-21,TLR2,TLR4 and NF-κB mRNA were detected by RT-PCR.Then the correlation between microRNA-21 content and TLR2,TLR4 and NF-κB levels were analyzed by Pearson.Results Rats with hepatocellular carcinoma showed pathological characteristics,such as abnormal mental state,dietary activity,hair growth,body weight and so on.More rats died.HE staining showed that the liver tissue of hepatocellular carcinoma rats was edema,abnormal color,rough surface,scattered ecchymosis,blunt edge,hard texture and easy to rupture.Under light microscope,the structure of hepatic lobules was disordered,hepatocytes in hepatic lobules atrophied,a large number of necrosis and liquefaction,and neutrophils infiltrated.RT-PCR showed that the microRNA-21 content in the model group was significantly higher than that in the control group.The expression levels of TLR2,TLR4 and NF-κB mRNA in the model group were significantly higher than those in the control group;microRNA-21 content in HCC cells was positively correlated with TLR2,TLR4 and NF-κB levels.Conclusion The expression of microRNA-21,TLR2,TLR4 and NF-κB is up-regulated in hepatocellular carcinoma rat cells,and the content of microRNA-21 is positively correlated with the levels of TLR2,TLR4 and NF-κB.
