参苏饮对“肺气虚外感”大鼠肺组织IL-1β和IL-18影响实验研究 被引量:6

摘要 目的:研究参苏饮对"肺气虚外感"大鼠肺组织中IL-1β和IL-18含量的影响及肺气虚外感证与NLRP3炎症小体的相关性。方法:将动物按体重随机分为空白组、模型组、参苏饮高剂量组、中剂量组、低剂量组,除空白组外,其余各组均采用"烟熏联合气管注射脂多糖(LPS)加冷风刺激"方法复制"肺气虚外感"证大鼠模型。酶联免疫吸附法(ELISA法)测定大鼠肺组织中IL-1β和IL-18的含量。结果:与空白组比较,模型组大鼠肺组织匀浆中IL-1β含量显著升高(P<0.05);与空白组比较,模型组、参苏饮高剂量组大鼠肺组织匀浆中IL-18含量显著升高(P<0.05);与模型组相比,参苏饮组大鼠肺组织匀浆中IL-1β含量显著降低(P<0.05),且参苏饮高、中、低不同剂量组之间肺组织匀浆中IL-1β含量无显著差异(P>0.05);与模型组相比,参苏饮中、低剂量组大鼠肺组织匀浆中IL-18含量显著降低(P<0.05),其中低剂量组大鼠肺组织匀浆中IL-18含量明显高于中剂量组(P<0.05),参苏饮高剂量组大鼠肺组织匀浆中IL-18含量无显著差异(P>0.05)。结论:NLRP3炎症小体下游炎症因子IL-1β、IL-18很可能参与"肺气虚外感"大鼠模型的炎症病理过程;参苏饮对"肺气虚外感"大鼠病理过程中IL-1β、IL-18的分泌可能起到抑制作用。"肺气虚外感"证病理过程的发生与NLRP3炎症小体存在一定相关性。 Objective:To Explore the influence of Shensu Drink on Interleukin-1βand Interleukin-18 in lung tissue of Lung-Qi exogenous model of rats;To study on the relationship between Exterior syndrome due to lung Qi-deficiency and NLRP3 inflammasome.Methods:The animals were randomLy divided into the blank control group,the model group,the high-dose group/middledose group/low-dose group of Shensuyin by weight,except the blank control group,other groups were built the Lung-Qi exogenous model of by fumigation and injecting lipopolysaccharide(LPS)through caudal veil and blowing cold wind.The content of IL-1β,IL-18 in lung tissue were determined with Enzyme-linked immunosorbent assay(ELISA).Results:Compared with the blank control group,the content of IL-1βin lung tissue homogenate in the model group were significantly increased(P〈0.05);The content of IL-18 in lung tissue homogenate in the model group and the high-dose group of Shensuyin were significantly increased(P〈0.05);Compared with the model group,the content of IL-1βin lung tissue homogenate of Shensuyin group were significantly decreased(P〈0.05);The content of IL-1βbetween Shensuyin high,middle and low group had no significant difference(P〉0.05);The content of IL-18 in lung tissue homogenate in middle-dose group/low-dose group of Shensuyin were significantly decreased(P〈0.05);Besides,the content of IL-18 in low-dose group of Shensuyin significantly higher than the middle-dose group Shensuyin(P〈0.05);The content of IL-18 between the model group and high-dose group of Shensuyin had no significant difference(P〉0.05).Conclusion:Inflammatory factor IL-1β,IL-18 actived by NLRP3 inflammasome may likely participated in the inflammatory process in LungQi exogenous model of rats;Shensuyin may couLd inhibit the secretion of IL-1β,IL-18 in the inflammatory process in Lung-Qi exogenous model of rats.The inflammatory process in exterior syndrome due to lung Qi-deficiency and NLRP3 inflammasome has a certain correlation.
