摘要
目的检测白细胞介素-1β(IL-1β)在类青光眼引流阀植入术后瘢痕组织中的表达水平,明确其在瘢痕形成中的作用。方法将30只新西兰大白兔随机分为硅胶组和新材料组,分别于球结膜下植入硅胶和聚一氯对二苯(Parylene C)涂层包裹的聚甲基丙烯酸甲酯(PMMA);于术后1、4、8周取球结膜至巩膜全层组织,采用苏木精-伊红(HE)染色检测组织中炎细胞浸润和成纤维细胞增生情况,于高倍镜下计数成纤维细胞;免疫组化检测IL-1β蛋白的表达及组织中的定位;Western blot法检测IL-1β蛋白表达水平;并统计分析IL-1β蛋白与成纤维细胞的相关系数。结果 HE染色显示:硅胶组炎细胞浸润、成纤维细胞增生数量明显高于新材料组;Western blot及免疫组化检测术后1、4及8周硅胶组的IL-1β蛋白的表达明显高于新材料组(P<0.05);免疫组化显示:IL-1β主要位于近巩膜面增殖的成纤维细胞周围。相关性分析结果表明:4、8周组织中IL-1β的表达水平与成纤维细胞呈正相关性(P<0.05)。结论 IL-1β可能在青光眼引流阀植入术后的瘢痕化形成中起促进作用。
Objective To observe interleukin-1β( IL-1β) expression on scarring after implantation of glaucoma drainage valve-alike,and to explore its effect on scarring. Methods Thirty New Zealand white rabbits were randomly assigned tosilicone group and new material group with 15 rabbits in each group,which implanted silicone and PMMA( Polymethyl methacrylate) coated Parylene C respectively under the conjunctiva. In the two groups,the whole tissues from conjunctiva to sclera were collected at 1 week,4 week and 8 week respectively after surgery.The expression of inflammatory cell infiltration and fibroblast proliferation in each group were observed by HE staining,and fibroblasts were counted by advanced microscope. Western blot and immunohistochemistry were used to detect IL-1β expression and location in tissues. The correlation coefficient was statistical analyzed between IL-1βprotein and fibroblasts. Results According to HE straining result,silicone group showed significantly higher expression of inflammatory cell infiltration and fibroblast proliferation compared with the new material group. According to Western blot and immunohistochemistry results,the concentrations of IL-1β were higher in silicone group than that in new material group at each time point( P 〈 0. 05). According to immunohistochemistry result,increased IL-1β was located around the proliferated fibroblasts on surface of the sclera. Correlation coefficient showed that the expression of IL-1β was positively correlated with the fibroblasts in 4 week and 8 week tissues( P 〈 0. 05).Conclusion IL-1β may promote the formation of scars after implantation of glaucoma drainage valve.
