摘要
目的:观察积雪草对人卵巢癌细胞A2780凋亡的影响并探讨相关机制。方法:用10μmol·L^(-1)、25μmol·L^(-1)、50μmol·L^(-1)、75μmol·L^(-1)的积雪草处理人卵巢癌细胞A2780 24 h,用MTT法检测细胞活性;用流式细胞术检测卵巢癌细胞A2780凋亡;用qRT-PCR检测卵巢癌细胞A2780中Bax、Bcl-2 mRNA的表达;用western blot法检测卵巢癌细胞A2780中Bax、Bcl-2蛋白的表达。结果:50μmol·L^(-1)、75μmol·L^(-1)的积雪草作用于细胞后,乳腺癌细胞A2780细胞形态发生明显改变,细胞皱缩、变圆。与对照组比较,75μmol·L^(-1)积雪草对卵巢癌细胞A2780抑制效果最明显。与对照组比较,50μmol·L^(-1)、75μmol·L^(-1)积雪草能够有效升高Bcl-2 mRNA的表达,降低Bax mRNA的表达,差异有统计学意义(P<0.01)。不同浓度积雪草均能有效升高Bax的表达并降低Bcl-2的表达,与对照组比较,差异有统计学意义(P<0.05)。结论:积雪草可抑制肿瘤的生长,其作用可能是通过诱导卵巢癌细胞A2780凋亡来实现的。
Objective: To observe the effect of herba centellae on apoptosis of human ovarian cancer cell line A 2780 and to explore the related mechanism. Methods: The human ovarian cancer cell A2780 was treated with 10 μmol · L-1,25 μmol·L-1,50μmol·L-1,75 μmol·L-1 of herba centellae for 24 h,and the cell activity was detected by MTT method. Apoptosis of ovarian cancer cell line A2780 was detected by flow cytometry. The expression of Bax,Bcl-2 mRNA in ovarian cancer cell line A2780 was detected by qRT-PCR. Western blot was used to detect the expression of Bax,Bcl-2 protein in ovarian cancer cell line A2780. Results: The morphology of breast cancer cell A2780 was obviously changed after treated with 50 μmol·L-1 and 75 μmol·L-1 of herba centellae,and the cell became round and shrinked. Compared with the control group,the inhibitory effect of A2780 was the most obvious effect with 75 μmol·L-1 of herba centellae,which was positively correlated with the concentration of drug. Compared with the control group,50 μmol·L-1 and 75 μmol·L-1 of herba centellae could effectively increase the expression of Bcl-2 mRNA and decrease the expression of Bax mRNA. The difference was statistically significant( P〈 0. 01). Different concentrations of herba centellae could effectively increase the expression of Bax and decrease the expression of Bcl-2,compared with the control group,the difference was statistically significant( P 〈0. 05). Conclusion: Herba centellae can inhibit tumor growth,which may be achieved by inducing apoptosis of ovarian cancer cell line A2780.
