依赖解旋酶的等温扩增技术在手足口病检测中的应用研究 被引量:2

摘要 目的探讨依赖解旋酶的等温扩增技术在手足口病检测中的应用研究。方法选取医院收治的手足口病儿童(EV71、CA16及HFMD其他EV病原体各40例)标本。对收集手足口病病人的标本使用依赖解旋酶恒温扩增技术检测,分析依赖解旋酶的恒温扩增(RT-t HDA)技术的特异性、灵敏度和符合率进行分析。结果检测均出现特异性扩增产物,具有典型的特异条带,检测标本病毒株均呈阳性,具有较强的检测特异性。阳性检测结果的符合率为91.28%~100.00%,平均为95.24%,阴性检测结果符合率为100.00%。结论依赖解旋酶恒温扩增技术具有快速、特异、简便、经济适用性高等优势,适用于基层医疗机构临床检测,值得临床借鉴与应用。 Objective To investigate the application of isothermal amplification technique based on helicase in the detection of hand-foot-mouth disease. Methods A sample of children with hand, foot and mouth disease(EV71, CA16 and HFMD other EV pathogens, 40 cases each) admitted to the hospital was selected. The specimens collected from patients with hand-foot-mouth disease were determined using the helicase thermostat amplification technique, and the specificity, sensitivity and coincidence rate of thermostat amplification(RT-t HDA) technology relying on helicase were analyzed. Results The detection products all showed specific amplification products with typical specific bands. The test sample virus strains were positive and had strong detection specificity. The coincidence rate of positive test results was 91.28% to 100.00% with an average of 95.24%, and the negative test result was 100.00%. Conclusion The thermocouple amplification-dependent helicase technology has the advantages of rapidity, specificity, simplicity, and high economic applicability. It can be applied to the clinical detection of primary medical institutions and the value of clinical reference and application.
