摘要
比较常规碱液提取、微波辅助碱液提取、超声辅助碱液提取麦麸蛋白,通过考马斯亮蓝法测定麦麸蛋白产率结果是:微波辅助与常规碱液提取麦麸蛋白得率一样;超声辅助碱液提取麦麸蛋白得率达到44.55%,是常规碱液法的1.30倍。通过凝胶电泳、红外光谱、热重分析测定麦麸蛋白的结构,结果显示:三种提取方法得到的麦麸蛋白分子量主要分布范围均为45 000~66 200和66 200~98 000,但超声辅助提取麦麸蛋白的α螺旋与β结构的比例降低并且该蛋白质的热重分析图在450℃时无失重峰,显示出较高的热稳定性及蛋白纯度。结果表明,超声辅助提取是获得具有分子灵活性的高产蛋白质的有效方法。为提高麦麸蛋白的产量,应用响应面分析法获得最佳提取条件,提取率达到68.04%,是常规碱液提取法的2倍。
Through the comparison of the investigation from the wheat bran protein(WBP) obtained from conventional alkali extraction(AE),microwave-assisted alkali extraction(MAE) and ultrasoundassisted alkali extraction(UAE),and according to the method of coomassie brilliant,the production rate of the wheat bran protein revealed that the yield of MAE-WBP was the same as AE-WBP and UAE method possess a higher extraction rate of 44.55%,which was about 1.30-fold than that of AE-WBP. The patterns of WBP obtained from the three extraction methods was distributed in the range of 45 000 ~ 66 200 and 66 200 ~ 98 000. However,the secondary structure of the protein produced by UAE did change with a reduce in the ratio of α-helix to β-structure by FT-IR analysis. Besides,the thermogravimetric analysis of the UAE-WBP showed no weight loss peak at 450 ℃,indicating its high thermal stability and protein purity. The results demonstrated that UAE is an effective method to acquire high-yield protein with excellent molecule flexibility. To improve the yield of the protein,response surface methodology was applied to obtain the optimum processing conditions,with extraction rate 68.04%,2 times as the yield of AE-WBP.
