摘要
目的:建立高效液相色谱法(HPLC)同时测定愈银片中5种指标性成分的含量。方法:色谱柱为Inertsil ODS-3(250mm×4.6 mm,5μm);乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱,检测波长为230 nm,柱温为30℃,流速为1.0 mL·min^(-1)。结果:芍药苷、芦丁、1,2,3,4,6-五没食子酰葡萄糖(PGG)、槲皮素和丹皮酚分别在0.132 3~1.323 0μg(r=0.999 8),2.001 6~20.016 0μg(r=0.999 8),0.076 4~0.764 0μg(r=0.999 8),0.050 86~0.508 6μg(r=0.999 8),0.112 28~1.122 8μg(r=0.999 8)线性关系良好,平均回收率分别为100.37%,100.71%,99.29%,100.81%,100.37%。结论:所建立的方法准确度高、重复性好、稳定可控,可为有效控制愈银片的质量提供依据。
OBJECTIVE To develop an HPLC method for the simultaneous determination of five index components in Yuyin tablets.METHODS The determination was performed on an Inertsil ODS-3（250 mm×4.6 mm,5μm）column with gradient elution using acetonitrile and0.1% phosphoric acid as the mobile phase.The detection wavelength was set at 230 nm,the column temperature was kept at 30 ℃,and the flow rate was 1.0 mL·^（-1）.RESULTS Paeoniflorin,rutin,1,2,3,4,6-O-pentagalloylglucose,quercetin and paeonol showed good linearity in the range of 0.132 3–1.323 0μg（r=0.999 8）,2.001 6–20.016 0μg（r=0.999 8）,0.076 4–0.764 0μg（r=0.999 8）,0.050 86–0.508 6μg（r=0.999 8）,0.112 28–1.122 8μg（r=0.999 8）,respectively.The average recoveries of the five index compounds were 100.37%,100.71%,99.29%,100.81% and100.37%,respectively.CONCLUSION This method is accurate,reproducible,stable and controllable,which provides the basis for the quality control of Yuyin Tablets.
