摘要
为了探究绵羊CD9基因g.208082881C>A位点多态性与产羔数之间的关系,利用Sequenom Mass ARRAYR~SNP技术对多羔绵羊品种(小尾寒羊380只)和单羔绵羊品种(滩羊、苏尼特羊、萨福克羊、杜泊羊和草原藏羊共380只)CD9基因g.208082881C>A位点多态性进行了检测,并与产羔数进行关联分析。结果表明:CD9基因g.208082881C>A位点存在AA和CA两种基因型,A基因为优势等位基因,等位基因频率在单羔、多羔绵羊品种间差异达到极显著水平(P<0.01);群体遗传学分析表明,该位点在杜泊羊群体中表现为低度多态(PIC<0.25),而在其他绵羊品种中均为中度多态(0.25<PIC<0.5);卡方适合性检验表明,该位点仅在杜泊羊群体中处于Hardy-Weinberg平衡状态(P>0.05);关联分析表明,该位点多态性与小尾寒羊不同胎次产羔数之间并无显著关联(P>0.05),AA型第二、三胎产羔数高于CA型。综上可知,CD9基因g.208082881C>A位点多态与小尾寒羊产羔数没有显著相关性。
To elucidate the polymorphism of sheep CD9 gene g.208082881 C>A locus and its association with the litter size,multiparous(380 Small-tail han sheep)and uniparous(total 380 for Tan,Sunite,Suffolk,Dorper and Prairie Tibetan sheep)sheep breeds were selected and Sequenom Mass ARRAYRSNPassay was applied to genotype g.208082881 C>A locus of CD9 gene. Then the association between the locus and the litter size in Small-tail han sheep was analyzed. From the result of genotyping,two genotypes(AA and CA)were found at g.208082881 C>A locus of CD9 gene in both uniparous and multiparous sheep,allele frequency was significantly different between uniparous and multiparous sheep(P<0.01). Population genetic analysis showed that the locus was at low polymorphism(PIC<0.25) in Dorper sheep,and at moderate polymorphism(0.25<PIC<0.5)in other sheep breeds. The χ2 test indicated that the locus of only Dorpersheep was under Hardy-Weinberg equilibrium(P >0.05). Association analysis indicated that the polymorphism of the locus of CD9 gene had no correlation with the litter size among the first,second or third parity in Small-tail han sheep(P >0.05),while the litter size of AA was higher than that of CA in the second and the third parity. Therefore,it could be concluded that polymorphism of the g.208082881 C>A locus of CD9 gene had no significant correlation with the litter size in Small-tail han sheep.
