摘要
目的 :优化超声-回流联用提取黄连-白术药对中主要有效成分小檗碱型生物碱的条件。方法 :以乙醇为溶剂,利用响应面分析法考察超声提取温度、超声功率、超声时间、乙醇浓度、黄连与乙醇料液比例、回流提取次数对黄连-白术药对中小檗碱型生物碱提取效果的影响。结果 :得出其最佳提取条件为将黄连、白术细粉浸泡于1∶35倍量的60%的乙醇中,置于47℃、40%功率下的恒温超声仪中提取40 min,再回流提取3次,得到的提取液小檗碱型生物碱提取率最高(30.96%)。结论 :得到的多元回归模型的准确性和适用性较好,可以用该模型来分析和预测黄连-白术药对中小檗碱型生物碱的超声-回流提取工艺。
Objective :To study the optimal conditions of berberine with ultrasonic-reflux extraction from Rhizoma Coptidis and Atractylodes. Methods :Alcohol was used as dissolvent. Response surface method(RSM)was used to study effects of ultrasonic temperature,ultrasonic power,ultrasonic time,ethanol concentration,solid-liquid ratio,reflux extraction times on the extraction yield of berberine. Results :The optimal conditions of berberine were as follows :solid-liquid ratio 1∶35,ethanol concentration 60%,ultrasonic temperature 47 ℃,ultrasonic power 40%,ultrasonic time 40 min,reflux extraction frequency 3 times. Under the optimal conditions,the extraction rate of berberine reaches 30.96%. Conclusion :Then the quadric polynomial regression model was established,and can be used to analyze and predict the extraction process of berberine from Rhizoma Coptidis and Atractylodes with ultrasonic-reflux extraction.
