摘要
AGL6基因在植物花器官的发育和花分生组织的形成中起着重要作用。为给探究AGL6基因在枣花发育过程中的功能奠定基础,作者基于其前期研究得到的枣转录组数据,采用RACE技术,从中秋酥脆枣中克隆得到了ZjAGL6基因的全长cDNA序列,并用实时荧光定量PCR技术分析了ZjAGL6基因的表达情况。序列分析结果表明,ZjAGL6基因编码区的序列长度为750 bp,共编码250个氨基酸;ZjAGL6具有保守的MADS-box和K-box两个结构域,该基因属于MADS-box基因家族中的TypeⅡ基因。系统进化分析发现,枣ZjAGL6与苹果MdAGL6的亲缘关系最近。实时荧光定量PCR分析结果表明,ZjAGL6基因在枣花和枣果中均有表达,但在枣吊、枣嫩叶和枣根中几乎不表达;在枣花发育过程中大量表达,其在大蕾期的表达量最高。
AGL6 gene plays an important role in development of floral organs and floral meristems. In order to lay an foundation for uncovering detailed role of ZjAGL6 during development of Ziziphusjujuba flowers, based on Z jujuba transeriptome data obtained in previous studies, a total length eDNA sequence of ZjAGL6 in Z jujuba 'Zhongqiusucui' was cloned by using RACE method, and expression of ZjAGL6 gene analyzed by using real-time PCR technology. The results of sequence analysis showed that length of coding region sequences of ZjA GL6 was 750 bp, encoding 250 amino acids. The results of protein structure prediction showed that ZjAGL6 display MADS-box domain and K-box domain, and the gene belonged to Type II gene in MADS-box gene family. The results of protein multiple sequence alignment and phylogenetic analysis showed that ZjAGL6 had a closer relationship with MdAGL6 ofMalus domestica. The results of real-time PCR analysis indicated that ZjAGL6 gene was expressed in both Z jujuba flowers and Z. jujuba fruits, but nearly no transcription detected in fruit bearing branch, tender leaves and roots; expression of ZjAGL6 was higher during flower development and reached the highest at big flower bud stage.
