摘要
蔗糖合成酶是蔗糖代谢的关键酶之一,它在调控果实品质和产量方面均有重大意义。为了探究ZjSS6基因在枣蔗糖合成代谢途径中的功能,文中基于枣的转录组测序数据,以中秋酥脆枣为试验材料,利用RACE技术克隆了枣SS6基因的全长cDNA序列,编码区长度为2 553 bp,编码850个氨基酸;该蛋白质的分子量为96.67 kDa,理论等电点为7.57。蛋白结构预测发现,该基因编码的蛋白含有蔗糖合成酶和糖基转移酶两个结构域。多序列比对和系统进化分析结果均表明,枣SS6与桑SS6的亲缘关系最近,其次是梅。实时荧光定量PCR表达分析结果显示,ZjSS6基因在枣各组织器官(果实、枣花、枣吊、叶片和根)中均有表达,其在幼果期枣果中的表达量最高,其次是绿果期枣果和枣吊,而在果实发育过程中其表达量呈整体下降趋势。
Sucrose synthase is one of the key enzymes of sucrose metabolism, and has great significance in regulation of fruit quality and yield. In order to explore the detailed function of ZjSS6 in sucrose metabolic pathways of Ziziphus jujube, based on Z. jujube transcriptome data, using Z. jujube 'Zhongqiusucui' as materials, and the full-length eDNA sequence ofZjSS6 gene was cloned by using RACE method, length of its coding sequences was 2 553 bp and it encoded 850 amino acids. Molecular mass and isoelectric point of the protein were 96.67 kDa and 7.57, respectively. Prediction results of protein structure showed that the protein coded by ZjSS6 contained two domain: sucrose synthase domain (SS domain) and glycosyl transferase group 1 domain (GT1 domain). The results of protein multiple sequence alignment and phylogenetic analysis showed that ZjSS6 had the closest relationship with the SS6 in Morus notabilis, and the next was in Prunus mume. The RT-PCR results indicated that ZjSS6 gene was expressed at all tissues (fruit, flower, shedding shoot, leaf and root), expression level was the highest in fruits at the young fruit stage, the next were in fruits and shedding shoots at the green fruit stage, but the overall expression level of ZjSS6 was trending downward during fruit development.
