肠道白色念珠菌感染对脾虚小鼠小肠黏膜中T细胞亚群和穿孔素-颗粒酶B的影响 被引量:1

摘要 目的:通过观察肠道白色念珠菌感染对脾虚小鼠小肠黏膜固有层中T细胞亚群和穿孔素、颗粒酶的影响,探讨机体在脾虚状态下感染白色念珠菌的部分免疫机制。方法:本实验采用饮食失节加劳倦过度的复合因素制备小鼠脾虚模型,经肠道感染白色念珠菌后第14天,应用流式细胞术检测小鼠小肠黏膜固有层中CD_3^+、CD_4^+和CD_8^+T的细胞百分比,采用免疫荧光法检测各组小鼠小肠黏膜中穿孔素、颗粒酶阳性表达细胞数。结果:与正常对照组比较,其他3组小鼠小肠黏膜固有层中CD_4^+T细胞比例均有不同程度下降,CD_8^+T细胞比例均有不同程度上升,有统计学意义(P<0.01);正常感染白念组和脾虚感染白念组小鼠小肠黏膜固有层中穿孔素阳性表达细胞均明显增多(P<0.05或P<0.01),而颗粒酶阳性表达细胞仅脾虚感染白念组明显增多(P<0.01)。其中,脾虚感染白念组最显著,与其他3组均具有统计学意义(P<0.05或P<0.01)。结论:小肠黏膜固有层中穿孔素和颗粒酶的高表达可能是造成小肠局部损伤的免疫机制之一,脾虚小鼠肠道局部免疫功能下降,感染发生后可加重肠道局部损伤程度。 Objective:To observe the effect of intestinal Candida albicans infection on T cell subsets and perforin-granzyme in small intestine lamina propria in the mice with spleen deficiency to investigate the immunologic mechanism of Candida infection in spleen deficiency condition.Methods:The compound factors of improper diet and overfatiguen were applied to establish a mouse model of spleen deficiency and infected by the intestinal Candida albicans.After 14 days,the percentage of CD3+,CD4+ and CD8+T in the lamina propria of small intestinal mucosa of mice was detected with flow cytometry and the expression of perforin and granzyme positive cells in small intestinal mucosa of mice was detected with immunofluorescence staining.Results:Compared with the normal control group,the proportion of CD4+T cells in the lamina propria of the intestinal mucosa in the other three groups decreased in different degrees and the proportion of CD8+T cells increased in different degrees,which was statistically significant(P〈0.01).The expression of perforin positive cells in the lamina propria of small intestinal mucosa of mice in the group infected by Candida albicans with no spleen-deficiency and with spleen deficiency were significantly increased(P〈0.05 or P〈0.01),and the positive expression of granzyme was only increased in group with spleen deficiency(P〈0.01).Among them,the infection of the spleen deficiency group was the most significant,and also statistically significant(P〈0.05 or P〈0.01) compared to the other 3 groups.Conclusion:The high expression of perforin and granzyme in the small intestinal mucosa lamina propria may be one of the immune mechanisms of local injury of small intestine.The intestinal immune function in mice with spleen deficiency was decreased and the degree of intestinal injury was increased after infection.
