摘要
目的:观察电针双侧"足三里"穴对实验性脾气虚大鼠小肠及下丘脑组织胃促生长素(Ghrelin)和血管活性肠肽(VIP)表达的影响,探讨对脾气虚证大鼠的治疗机制。方法:选用健康雄性SD大鼠40只,随机分为4组。其中正常对照组10只,气虚模型组10只,针刺治疗组10只,非经非穴组10只。除正常对照组外,其余3组均采用游泳力竭和饮食不节的复合因素建立脾气虚证模型。电针双侧"足三里"穴治疗6 d后,用10%水合氯醛腹腔麻醉(3 mL/kg体质量)。开腹,取小肠、下丘脑组织,液氮浸泡后冻存于-80℃冰箱待用;用酶联免疫吸附法(ELISA)检测大鼠小肠组织Ghrelin和VIP蛋白含量变化,荧光定量PCR法检测大鼠小肠及下丘脑组织Ghrelin、VIP及其受体mRNA表达的变化。结果:与正常对照组比较,气虚模型组、非经非穴组大鼠下丘脑和小肠组织中Ghrelin及其受体mRNA、VIP及其受体mRNA、Ghrelin和VIP蛋白表达均明显降低(P<0.05);与气虚模型组相比,针刺治疗组下丘脑和小肠组织Ghrelin及其受体mRNA、VIP及其受体mRNA、Ghrelin和VIP蛋白表达明显上调(P<0.05),非经非穴组未见显著性差异(P<0.05)。结论:针刺"足三里"穴通过调节脾气虚大鼠脑肠肽Ghrelin及VIP的表达水平,从能量代谢角度发挥对脾气虚大鼠的调控作用。
Objective:To observe the effects of gastric promoting growth hormone(Ghrelin) and vasoactive intestinal peptide(VIP) expressions in the small intestine and hypothalamus of electro-acupuncture at bilateral " Zusanli of the spleen qi deficiency rats and explore the therapeutic mechanism of spleen qi deficiency syndrome in rats.Method:Forty healthy SD rats were randomly divided into 4 groups:the normal group,spleen qi deficiency model group,acupuncture treatment group and non-acupoint group,10 rats in each group.In addition to the normal control group,the other three groups were established by combining way of swimming exhaustion and inappropriate diet to establish the model of spleen qi deficiency syndrome.After 6 days electro-acupuncture of Zusanli treatment,use 10% chloral hydrate intraperitoneal anesthesia(3 mL/kg body weight).Take out the small intestine and hypothalamus after laparotomy,and then soak in liquid nitrogen.Take it out and put it in the refrigerator for 80 degrees below zero and set aside.The changes of Ghrelin and VIP protein in small intestine were detected by enzyme linked immunosorbent assay.The expressions of Ghrelin,VIP and its receptor mRNA in small intestine and hypothalamus were detected by fluorescence quantitative PCR method.Results:Compared with the normal control group,Ghrelin and its receptor mRNA,VIP and receptor mRNA,Ghrelin and VIP protein expressions decreased obviously in model of spleen qi deficiency rats small intestine and hypothalamus.Compared with spleen qi deficiency model group,acupuncture treatment group’s Ghrelin and receptor mRNA,VIP and receptor mRNA,Ghrelin and VIP protein expressions significantly increased in small intestine and hypothalamic(P〈0.05).Conclusion:Electro-acupuncture of Zusanli plays a therapeutic role by regulating the expression level of brain-gut peptide Ghrelin and VIP in spleen deficiency rat from the perspective of energy metabolism.
