摘要
为建立一种快速、特异、敏感的鸡奇异变形杆菌检测方法。本研究针对鸡奇异变形杆菌全基因序列设计了1对特异性引物,建立检测鸡奇异变形杆菌的SYBR GreenⅠLC-PCR检测方法,进行敏感性、特异性、重复性试验,并通过临床组织样品检测评价其临床实用性。结果显示,所建立方法的标准曲线线性关系良好,相关系数R^2=0.9957。该方法灵敏度高,可检测到稀释度为10~1 CFU/m L的奇异变形杆菌。特异性强,不与其他禽源致病原发生交叉反应。重复性试验变异系数小于2%。提示该方法可用于鸡奇异变形杆菌的监测和早期临床诊断。
For establishment of a rapid,specific and sensitive detection method for Proteus mirabilis in chicken,we designed a pair of specific primers for r DNA sequence of the bacteria,established the SYBR GreenⅠLC-PCR,tested its sensitivity,specificity and repeatability,and evaluated its clinical utility by detecting the clinical tissue samples.The standard curve revealed a good linear relationship with the correlation coefficient R^2=0.995 7.The method showed high sensitivity,with the minimum detected dilution of 10^1CFU/mL for Proteus mirabilis,high specificity,no cross-reactons with other avian pathogens,and well repeatability with the coefficient of variation less than 2%.The results suggest the established technique could be used for monitoring and early diagnosis of Proteus mirabilis in chicken.
