摘要
研究旨在鉴别检测鸡场中大肠杆菌、沙门氏菌、空肠弯曲杆菌和奇异变形杆菌等4种常见病原菌。试验在已有4种病原菌单一定量PCR检测方法的基础上,采用SYBR GreenⅠ多重定量PCR检测方法检测大肠杆菌、沙门氏菌、空肠弯曲杆菌和奇异变形杆菌。结果显示,建立检测鸡大肠杆菌、沙门氏菌、空肠弯曲杆菌和奇异变形杆菌SYBR GreenⅠ多重定量PCR检测方法与其他菌株无交叉反应;大肠杆菌、沙门氏菌、空肠弯曲杆菌和奇异变形杆菌的最低检测限分别为5.6×10^(1)、8.9×10^(0)、7.8×10^(0)、3.8×10^(1)CFU/mL;Ct值变异系数在2.22%之内。研究表明,试验所建立的方法特异性强、灵敏性高、重复性好,可应用于临床中大肠杆菌、沙门氏菌、空肠弯曲杆菌和奇异变形杆菌感染的快速检测。
The aim of the study was to identify four common pathogens in chicken farms,including Escherichia coli,Salmonella,Campylobacter jejuni and Proteus mirabilis.The SYBR GreenⅠmultiplex quantitative PCR method was used to detect Escherichia coli,Salmonella,Campylobacter jejuni and Proteus mirabilis on the basis of the existing single quantitative PCR method.The results showed that the SYBR GreenⅠmultiplex quantitative PCR method for the detection of chicken Escherichia coli,Salmonella,Campylobacter jejuni and Proteus mirabilis had no cross-reaction with other strains.The lowest detection limits of Escherichia coli,Salmonella,Campylobacter jejuni and Proteus mirabilis were 5.6×10^(1)、8.9×10^(0)and7.8×10^(0)、3.8×10^(1)CFU/mL,respectively.The coefficient of variation of Ct value was within2.22%.The study indicates that the established method has high specificity,high sensitivity and good reproducibility,and can be applied to the rapid detection of Escherichia coli,Salmonella,Campylobacter jejuni and Proteus mirabilis infections in clinic.
