摘要
目的探讨银杏叶提取物(GBE)对肝纤维化模型大鼠的影响及抗肝纤维化机制。方法 SD大鼠60只随机分为正常组(n=12)和模型组(n=48),模型组大鼠又分为4个亚组:模型对照组、GBE低、中、高剂量组(n=12),GBE低、中、高剂量组每天分别灌胃给予GBE 50 mg/kg、100 mg/kg及200 mg/kg,正常组和模型组给予等体积的0.9%氯化钠溶液,连续2个月(1次/d)。给药结束后腹主动脉取血,分离血清,试剂盒检测肝功能指标(ALT、AST、MDA、SOD、白蛋白、球蛋白及白/球),放免法检测肝纤维化指标(HA、PC-Ⅲ、C-Ⅳ及LN),大鼠脱颈椎处死后立即取右叶部分肝组织,液氮速冻后于-80℃保存,RT-PCR检测肝组织TGF-β1、TIMP1、Ⅰ型胶原mRNA的表达水平。结果与模型对照组比较,GBE中、高剂量组AST、ALT、球蛋白及肝纤维化各项指标显著降低,TGF-β1、TIMP1、Ⅰ型胶原mRNA表达水平显著降低(P<0.05),白蛋白水平、白/球值显著升高(P<0.05),MDA水平差异无统计学意义(P>0.05)。结论 GBE能显著改善肝纤维化大鼠的肝功能指标,肝纤维化指标,其机制可能是通过降低TGF-β1、TIMP1、Ⅰ型胶原mRNA表达水平来实现的。
Objective To investigate the effects of extract of ginkgo biloba leaf (GBE) on liver fibrosis of rats,and to explore its action mechanism against liver fibrosis.Methods Sixty SD rats were randomly divided into normal control group (n =12) and model group (n =48).The rats in model group were redivided into four subgroups:model control group,low-dose GBE group,median-dose GBE group and high-dose GBE group,with 12 rats in each group.The rats in three GBE groups were given GBE 50mg/kg,100mg/kg,200mg/kg,respectively by gavage,however,the rats in normal control group and model control group were given equal volume of 0.9% sodium chloride solution,once a day for 2 months.After administration,the serum levels of ALT,AST,MDA,SOD,albumin,globulin (albumin/globulin) were detected by Kit.The liver fibrosis indexes including HA,PC-Ⅲ,C-Ⅳ and LN were measured by radioimmunoassay.After the rats were sacrificed,the right lobe of the liver tissue was taken out,and the expressin levels of TGF-β1,TIMP1,typeⅠcollagen mRNA were detected by RT-PCR.Results As compared with those in model control group,the serum levels of AST,ALT,globulin and hepatic fibrosis indexes in median-dose GBE group and high-dose GBE group were significantly decreased,and the expression levels of TGF-β1,TIMP1 and typeⅠcollagen mRNA were significantly decreased too (P〈0.05).However the levels of albumin and the ratio of albumin/globulus were significantly increased (P〈0.05),but the levels of MDA were not changed obviously.Conclusion The extract of ginkgo biloba leaf can significantly improve liver function indexes and hepatic fibrosis indexes of rats with liver fibrosis,and its action may be completed by reducing the expression level of TGF-β1,TIMP1 and type Ⅰcollagen mRNA.
