摘要
采用牛血清白蛋白免疫损伤大鼠肝纤维化模型 ,以虫草菌丝和秋水仙碱为对照 ,通过肝组织病理学与Ⅰ、Ⅲ、Ⅳ型胶原及TGFβ1 、TGFβR1 、Dm阳性细胞免疫组化观察 ,肝Hyp含量与肝胶原酶活性检测 ,观察虫草多糖抗肝纤维化的作用与机制。结果显示 ,虫草多糖与虫草菌丝及秋水仙碱均能减轻肝纤维化程度 ,降低Hyp含量 ,减少胶原及TGFβ1 生成与分泌 ,降低TGFβR1 表达 ,减少Dm阳性细胞 ,其中虫草多糖对Ⅳ型胶原与TGFβR1 表达的影响更为明显。提示虫草多糖具有良好的抗肝纤维化作用 ,可能是虫草菌丝抗肝纤维化的主要有效成分之一 ,其作用机制可能在于抑制肝星状细胞的活化及其TGFβ1 的表达。
The study was to investigate the actions of cordyceps polysaccharide on liver fibrosis and its related mechanisms.The liver fibrosis was induced through injecting bovine serum albumin in rats,and treated by cordyceps polysaccharide and cordyceps mycelium respectively,and with colchine as control.The hepatic pathological changes,type Ⅰ,Ⅲ,Ⅳ collagens,transforming growth factor β 1(TGFβ 1)and its receptor(TβR 1),and the cells expressing Dm were observed with immunohistochemistry method.The hepatic Hyp content and interstitial collagenase activity were measured.The results showed that all above drugs could attenuate liver fibrosis,decrease hepatic Hyp content and collagen production,reduce TGFβ 1 production and TβR 1 expression and decrease cell Dm expressing.It is suggested that cordyceps polysaccharide has good action against liver fibrosis,which is perhaps one of the main effective components from cordyceps mycelium.The mechanism of cordyceps polysaccharide action on liver fibrosis is associated with the inhibition of hepatic stellate cell activation and TGFβ 1 expression.
