摘要
目的研究胃部非癌组织、胃部癌旁组织与胃部癌组织中自噬基因Beclin1和微管相关蛋白轻链3(microtubule-associated protein 1 light chain 3,MAPLC3)基因的表达含量,确定Beclin1和MAPLC3表达含量与胃癌之间的联系。方法使用实时逆转录-聚合酶链反应(Real-Time reverse transcription polymerase chain reaction,Real-Time RTPCR)检测胃部非癌组织、胃部癌旁组织与胃部癌组织中Beclin1以及MAPLC3基因的mRNA表达含量。使用蛋白质印迹法(Western blot)方法检测胃部非癌组织、胃部癌旁组织与胃部癌组织中Beclin1以及MAPLC3基因的蛋白表达含量。结果与胃部非癌组织相比,胃部癌旁组织Beclin1以及MAPLC3的mRNA含量显著降低(P<0.01),胃部癌组织中Beclin1以及MAPLC3的mRNA含量也显著降低(P<0.01);与胃部癌旁组织相比,胃部癌组织中Beclin1以及MAPLC3的mRNA含量也显著降低(P<0.05)。与胃部非癌组织相比,胃部癌旁组织Beclin1以及MAPLC3的蛋白含量显著降低(P<0.05),胃部癌组织中Beclin1以及MAPLC3的蛋白含量也显著降低(P<0.01);与胃部癌旁组织相比,胃部癌组织中Beclin1以及MAPLC3的蛋白含量也显著降低(P<0.05)。结论自噬相关基因Beclin1以及MAPLC3的mRNA和蛋白水平在胃癌组织中表达降低,这提示自噬相关基因与胃癌的发生、发展密切相关。
Objective: To study the non stomach cancer tissue, autophagy gene Beclinl and microtubule stomach tissues and gastric cancer associated protein light chain 3 (microtubule-associated protein 1 light chain 3, MAPLC3) expression lev- els of genes, and determine Beclinl and MAPLC3 expression between gastric cancer and content contact. Methods: The real- time reverse transcription polymerase (Real-Time reverse transcription poly chain reaction Merase chain reaction, Real-Time RT-PCR) was used to detect gastric non cancer tissues, Beclinl stomach tissues and the stomach cancer tissue and MAPLC3 gene mRNA expression content. Using Western blotting ( Western blot) method was used for detection of gastric non cancer tissue the expression of Beclinl of gastric tissues and gastric cancer tissues, MAPLC3 protein content. Results: Compared with non stomach cancer, the mRNA contents of stomach cancer tissues and Beclinl MAPLC3 were significantly decreased (P 〈 0.01 ), stomach carcinoma Bechnl and MAPLC3 content of mRNA was lower (P 〈 0.01 ) ; and stomach carcinoma, stomach carcinoma Beclinl and MAPLC3 content of mRNA was lower (P 〈 0.05). Compared with the non stomach cancer tissue, stomach cancer adjacent tissue Beclinl and MAPLC3 protein content was significantly reduced (P 〈 0.05), gastric carcinoma Beclinl and MAPLC3 protein content also decreased significantly (P 〈0.01 ) ; Compared to noncancerous tissues, gastric carcinoma Beclinl and MAPLC3 protein content also decreased significantly (P 〈 0.05 ). Conclusion: The autophagy related gene Beclinl and MAPLC3 mRNA and protein expression decrease in gastric cancer tissues, suggesting that autophagy related genes are closely related with gastric carcinogenesis and development.
