摘要
目的了解诱导HO-1表达对慢性间歇性缺氧下INS-1细胞凋亡的影响。方法通过不同浓度CoPP作用于慢性间歇性缺氧下INS-1细胞,观察胰岛素水平、HO-1及Caspase-3水平,采用RT-PCR检测HO-1、Caspase-3mRNA表达。结果随着CoPP浓度增高,胰岛素、HO-1表达逐渐增高,而Caspase-3逐渐降低,而CoPP浓度达到40umol/L时作用最强(P<0.05)。结论慢性间歇性缺氧下诱导INS-1细胞HO-1表达,抑制凋亡相关蛋白Caspase-3,在一定程度上对细胞起保护作用。
Objective To understand the effect of HO-1 expression on the apoptosis of INS-1 cells induced by chronic intermittent hypoxia. Methods INS-1 cells were observed under different concentrations of CoPP in chro-nic intermittent hypoxia, and the levels of insulin were observed. It used RT-PCR to detect the expression of HO-1 and Caspase-3 mRNA. Results With the increase of CoPP concentration, the expression of insulin and HO-1 mRNA gradually increased, while Caspase-3 mRNA decreased. When the concentration of CoPP reached 40umol/L, the strongest effect was observed. Conclusion The induced expression of HO-1 in INS-1 cells can inhibit protein Caspase-3, and protected the cells at certain extent.
