摘要
目的探讨两种模式间歇低氧对大鼠血糖、胰岛素及胰腺组织Caspase-3活性及其表达的影响。方法将63只SD大鼠随机分为常氧对照组、间歇低氧1组(高频轻度)、间歇低氧2组(低频重度),3组又各自随机分为暴露1、2、4周组,每组7只。3个暴露4周组分别于0周及暴露1、2、4周检测空腹血糖;3组大鼠分别于暴露1、2、4周检测胰岛素水平,胰腺组织Caspase-3活性及其mRNA和蛋白表达水平。结果间歇低氧1组暴露4周组血糖从暴露2周开始升高,暴露4周与暴露2周比较差异无统计学意义(P>0.05);间歇低氧2组暴露4周组血糖从暴露1周即出现升高,随着暴露时间的延长进行性增高,暴露4周较间歇低氧1组暴露4周增高(P<0.05)。间歇低氧1组血清胰岛素水平呈缓慢上升趋势,暴露4周高于常氧对照组(P<0.01);间歇低氧2组血清胰岛素从暴露1周即开始增高,暴露2周达高峰,暴露4周下降至常氧对照组水平。间歇低氧2组胰腺组织Caspase-3活性及其mRNA和蛋白表达水平随暴露时间的延长呈进行性增高(P<0.05,P<0.01)。结论低频重度间歇低氧模式对胰腺组织的损害较高频轻度间歇低氧模式严重,且呈进行性加重。
Objective To study the effects of two different patterns of intermittent hypoxia on glucose,insulin,and activity and expression of pancreas caspase-3 in rats. Methods Sixty- three Sprague- Dawley rats were randomly divided into control group( n = 21),intermittent hypoxia group 1( group A,high frequence and mild degree,n = 21)and intermittent hypoxia group 2( group B,low frequence and severe degree,n = 21). The three groups were randomly divided into three subgroups,and were exposed for 1 week,2 weeks and 4 weeks respectively( n = 7 for each subgroup).Levels of fasting plasma glucose( FPG) in the rats were measured at 0 week,and exposure for 1 week,2 weeks and 4weeks,and then levels of insulin in serum,caspase- 3 mRNA,protein and activity of pancreas of all rats were detected after exposure for 1 week,2 weeks and 4 weeks. Results The group A was exposured for 4 weeks,and glucose level was increased after exposure for 2 weeks,but there was no significant difference in the level between exposure for 2 weeks and 4 weeks( P 〉0. 05); group B was exposured for 4 weeks,and glucose level was increased after exposure for 1 week,and glucose level was progressively increased with prolonging exposure time,and the level after exposure for 4 weeks was significantly higher than that in group A( P〈0. 05). Insulin level in group A showed a slowly upward tendency,and the level after exposure for 4 weeks was significantly higher than that in control group( P〈0. 01); in group B,the insulin level began to rise up after exposure for 1 week,and the peak emerged after exposure for 2 weeks,and then the difference in the level showed no statistical significance after exposure for 4 weeks between group B and control group( P〉 0. 05).The levels of Caspase-3 activity and mRNA in group B were progressively increased with prolonging time( P〈0. 05,P〈0. 01). Conclusion Low frequence and severe degree of intermittent hypoxia can cause severe damage on pancreas tissues than that by high frequence
