摘要
目的确定人参糖蛋白的基本化学构成,探讨人参糖蛋白抗β-淀粉样蛋白(Aβ25-35)引起的神经炎症和神经保护作用。方法采用超滤、葡聚糖凝胶等方法分离纯化得到人参糖蛋白(Pg Gp),采用苯酚-硫酸法测定总糖含量,Lowry法测定蛋白含量,高效液相色谱法测定纯度和分子量分布。利用MTT法考察Pg Gp(5,25,125μg/m L)对细胞存活率的影响;Griess法测定Pg Gp对Aβ25-35诱导的SH-SY5Y细胞一氧化氮释放的影响。结果Pg Gp总糖含量为25.9%,蛋白含量为73.4%,分子量分布在200~50 000 Da。Pg Gp在不影响细胞存活率情况下,中、高剂量组与模型组比较,表现出保护Aβ25-35诱导的SH-SY5Y释放NO的能力(P<0.05,P<0.01)。结论人参糖蛋白可降低Aβ25-35诱导SH-SY5Y细胞一氧化氮释放量,从而具有保护神经细胞的作用。
Objective To confirm chemical composition of Panax ginseng glycoprotein (PgGp). Discussion of PgGp anti-inflammatory effect of neuritis,which was caused by Aβ25-35 and the protective effect of PgGp on the nerves. Methods For separation and purification of PgGp, a hollow fiber ultrafiltration device was used. The content of total sugar was measured by the phenol sulfuric acid method. The content of protein was measured by Lowry method. The purity and molecular weight distribution was measured by HPLC and GPC. The influence effect of PgGp on cell viability is measured using the MTT. The influence on the SH-SY5Y cells,which was processde by Aβ25-35, release Nitric Oxide of the effect of PgGp is measured using the Griess method. Results The total sugar content of ginseng glycoprotein was 25.9%, the content of protein was 73.4%. The molecular weight was 200-50000 Da. Middle and high dose group showed significant activity of protective SH-SY5Y induced by Aβ25-35. Conclusion PgGp, which reduce the amount of Nitric Oxide on Aβ25-35 destrucitved SH-SY5Y cells, is nmning the protection of the safety of nerve cell.
