摘要
目的通过对人参TIFY基因家族的保守基序,系统进化,基因模式表达分析,深入研究TIFY基因功能,为人参优良品种选育提供理论依据。方法通过生物信息学手段对人参TIFY基因家族进行鉴定和分析,使用TAIR、ExPASy、WOLF PSORT、MEME、NCBI等在线网站以及TBtools、MEGA等软件进行TIFY基因表达模式分析,并进行可视化作图。结果在人参中共鉴定出33个TIFY基因成员,分别聚类为ZML、PPD、JAZ、TIFY 4个亚家族,其氨基酸数量在104~1064 aa,等电点介于4.96~9.78,分子质量在117477.88~49576.63 D之间,大部分被鉴定的基因定位于细胞核中,通过热图对基因的表达分析表明,人参TIFY基因家族在根、根茎、叶、花、果实中表达存在一定的差异性。结论通过对人参TIFY全基因组水平鉴定及生物信息学分析,推测TIFY基因家族在对人参的生长发育有重要的调控作用,并且可能参与了人参皂苷的生物合成,将为人参的优良品种选育提供理论依据。
Objective The conserved motifs,phylogenetic evolution and gene pattern expression of Renshen(Panax ginseng C.A.Meyer)TIFY gene family were analyzed.The study of TIFY gene function provides some basic theory for the selection and breeding excellent Renshen(Panax ginseng C.A.Meyer)varieties.Method The TIFY gene family was identified and analyzed by bio-informatics methods,and online websites such as TAIR,ExPASy,WOLF PSORT,MEME and NCBI,as well as softwares such as TBtools and MEGA were used to analyze the prediction and expressions of TIFY genes,and visualize the diagrams.Result There were 33 TIFY gene members in Renshen(Panax ginseng C.A.Meyer),clustered into four subfamilies,such as ZML,PPD,JAZ and TIFY.The number of amino acids was 104~1064 aa,the isoelectric point ranged from 4.96 to 9.78,the molecular weight was 117477.88~49576.63 D,and these proteins were almost uniformly localized in the nucleus.The analysis of gene expression pattern by heat map showed that there were some differences in the expression of Renshen(Panax ginseng C.A.Meyer)TIFY gene in the same root,rhizome,leaf,flower and fruit.Conclusion In the analysis of genomic level identification and bio-informatics of Renshen(Panax ginseng C.A.Meyer)TIFY,it is speculated that TIFY gene family plays an important role in the growth and development of Renshen(Panax ginseng C.A.Meyer),and may be involved in the biosynthesis of ginsenoside,which will provide a theoretical basis for the breeding of good varieties of Renshen(Panax ginseng C.A.Meyer).
