摘要
目的探讨防己黄芪汤治疗胰岛素抵抗的可能作用机制。方法将SD大鼠随机分为空白组、模型组、中药组及西药组各10只,除空白组外其余各组采用高脂饲料饲养12周建立胰岛素抵抗模型。造模成功后中药组按1 ml/100 g灌服防己黄芪汤(浓度0.6 g/ml),西药组按1 ml/100 g灌服吡格列酮悬浊液(浓度1 mg/ml),模型组及空白组灌服等容量蒸馏水,每日1次,各组连续给药2周。造模后检测各组大鼠葡萄糖输注率(GIR);给药后检测血清游离脂肪酸、脂联素、瘦素、抵抗素含量;油红O染色观察骨骼肌脂肪异位沉积程度;Western-blotting法测定骨骼肌腺苷酸活化蛋白激酶(AMPK)、p38丝裂原活化蛋白激酶(p38MAPK)蛋白含量。结果与空白组比较,模型组大鼠血清游离脂肪酸、瘦素、抵抗素含量及p38MAPK蛋白表达显著升高,脂联素含量、GIR、AMPK蛋白表达显著降低(P<0.05或P<0.01);与模型组比较,西药组、中药组游离脂肪酸、瘦素、抵抗素含量及p38MAPK蛋白表达显著降低,脂联素含量、AMPK蛋白表达显著升高(P<0.05或P<0.01)。油红O染色显示,与模型组比较,中药组和西药组大鼠骨骼肌红染颗粒减少,脂质沉积改善。结论防己黄芪汤可改善大鼠胰岛素抵抗脂质代谢紊乱,调节脂肪因子水平,其机制可能是通过调节AMPK信号转导通路蛋白水平实现的。
Objective To explore the potential mechanism of Fangji Huangqi Decoction(防己黄芪汤) in treating insulin resistance(IR). Methods SD rats were randomized into the blank group,the model group,Chinese medicine group and Western medicine group,with 10 rats in each group. Every group was given high-fat diet for 12 weeks to build IR model except the blank group. After modeling,Chinese medicine group was administered Fangji Huangqi Tang(0. 6 g/ml) by gavage at a dose of 1 ml/100 g,Western medicine was given Pioglitazone suspension(1mg/ml)by gavage at a dose of 1 ml/100 g and the model group and the blank group were given distilled water of equal volume.The drugs were given for continuous 2 weeks. After modeling,the glucose infusion rate(GIR) of each group was detected; after administration,the volume of free fatty acid(FFA),adiponectin(APN),leptin(LP) and resistin were detected; the ectopic fat deposition level in skeletal muscle was observed by oil red O staining; the volume of AMP-activated Protein Kinase(AMPK) and p38 mitogen-activated protein kinase(p38 MAPK) in skeletal muscle were detected by Western-blotting method. Results Compared with the blank group,the volume of FFA,LP and resistin and the expression of p38 MAPK in the model group were significantly higher,while the volume of APN,GIR and the expression of AMPK were significantly lower(P〈0. 05 or P〈0. 01). Compared with the model group,the volume of FFA,LP and resistin and the expression of p38 MAPK in Chinese medicine group and Western medicine group were significantly lower,while the volume of APN and the expression of AMPK were significantly higher(P〈0. 05 or P〈0. 01). The oil red O staining showed that,compared with the model group,the red stained particles in the skeletal muscle of Chinese medicine group and Western medicine group were significantly fewer,and the lipid deposition ameliorated. Conclusion Fangji Huangqi Decoction could improve the IR and the disturbance of lipid metabolism of rat
