摘要
为结合液体菌种与固体菌种的优点、克服液体菌种不能长距离运输的不足,对杏鲍菇液体培养的菌丝球与基质结合的再固化技术进行了研究。通过菌丝萌发力与生长速度的比较筛选出最适合"杏大"的基质配方为玉米芯78%,麦麸20%,石膏2%,与其他基质配方相比差异显著(P〈0.01)。25 d内保藏时间对菌丝的生长速率和菌丝萌发力没有明显影响,差异不显著(P〉0.05)。对优化的再固化菌种在不同的保藏时间进行呼吸强度和纤维素酶活测定,结果显示,再固化菌种呼吸强度随着保藏时间的增加而逐渐增加,25℃保藏35~45 d时菌丝的呼吸强度急剧上升,在45 d时达到最大1.132 8 mg/(g·h);纤维素酶活随保藏时间下降,菌种活力下降,因此再固化菌种的保藏时间在35 d为宜。
The study on resolidification spawn was to combine the advantages ot hquld spawn wltn solid spawn, and to overcome the shortcomings of liquid spawn which can not be long-distance transported. By comparing the growth rate and germination time of the mycelium at different preservation time, the optimum media component (78% corn cob , 20% wheat bran and 2% gypsum) were screened and there was a significant difference(P〈0.01 ) with other components, there were no significant difference among the preservation time of 5, 15 and 25 d (P〉0.05). The respiration intensity and cellulase activity of the optimum solidification spawns were determined by the germi- nation of the spawns at different preservation time, the results showed that the respiration intensity increased gradually with the increase of storage time, the respiration rate of mycelium goes up quickly at day 35 -45 with temperature of 25℃, and reached maximum 1. 1328 mg ( CO2 ) / g ( fresh weight) · h at 45 d, and cellulase activity started to slow down, the activity of the spawn al- so went down, so the preservation time should not go beyond 35 days.
