摘要
目的:明确激素性股骨头坏死(SANFH)大鼠模型形成过程中骨组织Ca、P、Mg、Zn元素含量的变化规律。方法:96只SD大鼠随机分为空白组与模型组,每组48只,采用尾静脉注射大肠杆菌脂多糖40μg/kg,2d后臀肌注射甲基强的松龙40mg/kg,3d,建立SANFH模型;空白组臀肌注射等体积0.9%氯化钠溶液。分别于造模4、8、12周后分批处死各组大鼠,分离股骨,通过系统方法学考察建立股骨中各元素电感耦合等离子体发射光谱法(ICP-OES)含量测定方法,分析比较SANFH大鼠模型形成过程中股骨各元素含量变化。结果:各元素在其线性范围内均呈现良好的线性关系(r≥0.999),平均加样回收率在99.96%-101.40%之间(RSD<3%,n=6)。与空白组比较,模型组股骨Ca、P元素含量均呈下降趋势,但雄性模型组大鼠股骨Ca、P含量在造模8、12周显著下降(P<0.05);雌性模型组大鼠股骨P含量在造模8周显著性下降(P<0.05);而模型组大鼠股骨Mg、Zn含量则无显著差异。结论:SANFH发病过程中会伴随引起骨组织Ca、P元素含量的变化,提示可将股骨Ca、P含量作为SANFH模型评价指标及药物干预的药效评价指标。
Objective:To study the relationship between steroid-incrced necrosis of femur head(SANFH) and the change of content of bone calcium,phosphorus,magnesium,and zinc.Methods:Ninty-six SD rats were randomly divided into control group and model group,with 48 rats in each group.The model was established by intra-muscular injection of 40mg/kg methylprednisolone for 3 days after intravenous injection of 40μg/kg LPS for 2 days.The control group was injected with equal volume of normal saline.Rats were sacrificed to harvest femurs at 4th,8th,12 th weeks,the content change of calcium,phosphorus,magnesium and zinc in SANFH rats were measured by ICP-OES method.Results:All elements showed a good linear relationship(r≥0.999),the average recovery rate was between 99.96%and 101.40%(RSD3%,n=6).Compared with control group,all the content of elements in model group were reduced,but the content of bone calcium and bone phosphorus was significantly reduced at 8th and 12 th week of the male rats of model group(P〈0.05),and the content of bone phosphorus was significantly reduced at8 th week of the female rats in model group(P0.05).However,there was no significant difference between control group and model group in the content of magnesium and zinc.Conclusion:During the process of SANFH,the content of bone calcium and phosphorus is changed,this can be used to evaluate the SANFH model and the efficacy of drugs.
