摘要
芡实壳为芡实种仁的外壳,是芡实加工过程中的废弃物。采用HPLC-DAD-MS/MS对芡实壳成分进行分析,首次鉴定了芡实壳中主要活性多酚成分——鞣花酸,建立了其HPLC定量分析方法。鞣花酸标准曲线线性关系为0.9994,HPLC色谱峰保留时间相对标准偏差为0.41%,峰面积相对标准偏差为1.14%,加标回收率为(100.40±3.14)%。以乙醇为提取溶剂,对微波辅助法提取芡实壳中鞣花酸的工艺条件进行单因素试验优化。结果表明:最佳提取工艺条件为:乙醇体积分数75%,微波功率600 W,微波时间4 min,料液比1∶50。在此条件下,测得芡实壳中鞣花酸含量为11.51 mg/g。与其它一些植物原料相比,芡实壳具有资源丰富,价格低廉,鞣花酸含量较高的特点,具有很好的开发应用前景。
Gordon euryale seed shells(GESS) is the by-product of Gordon euryale seed process. In the present study, the chemical constituents in GESS were analyzed by HPLC-DAD-MS/MS. Ellagic acid was identified for the first time and its HPLC quantitative method was developed. The correlation coefficients of ellagic acid calibration curve was0.9994, the RSD of tRand peak area were 0.41 and 1.14%, respectively, while the recovery was(100.40 ± 3.14)%. The microwave-assisted method for the extraction of ellagic acid from GESS was optimized. The results showed that the optimum extraction conditions were: ethanol concentration 75%, microwave power 600 w, microwave time 4 min, ratio of material and extracting solvent 1∶50(m/V, g/m L). Under the optimum extraction conditions, the content of ellagic acid in GESS was quantified as 11.51 mg/g.
