Determination of riboflavin content in chicken essence seasoning by high performance liquid chromatography

摘要 建立了反相高效液相色谱法快速测定鸡精中核黄素含量的方法。样品经热水提取后通过高效液相色谱分析,以色谱峰保留时间和紫外可见光谱进行定性。采用默克LiChrospher 100 RP-18e色谱柱,以甲醇-乙酸铵(0.02mol/L)为流动相,流速为1.0mL/min,检测波长为445nm,外标法测定核黄素含量。结果表明,核黄素在0.5 50mg/L范围内线性关系良好(R2=0.9996),检出限(S/N=3)为0.16mg/L。在3个浓度水平下加标回收率在87.1%102.0%之间,相对标准偏差(n=6)在0.73%2.93%之间。 A rapid-HPLC method for the determination of riboflavin in chicken essence seasoning was developed.The sample was extracted with hot water and chromatographied on a Merck LiChrospher 100 RP-18e column and detected at 445nm,with a mobile phase of methanol-ammonium acetate solution(0.02mol / L) at a flow rate of 1.0mL / min.The linearity was obtained in the range of 0.5 50mg / L(R2 = 0.9996).The determination limit(S / N = 3) was 0.16mg / L.The recovery of three different concentrations were in the range of 87.1% 102.0% and RSD’s(n = 6) was in the range of 0.73% 2.93%.
