Determination of Ellagic Acid in Strawberry by Dispersive Solid Phase Extraction-High Performance Liquid Chromatography

摘要 建立了分散固相萃取-高效液相色谱法(DSPE-HPLC)测定草莓中游离鞣花酸含量的检测方法。在酸性条件下用甲醇提取,经C18分散固相萃取净化。采用Agilent Poroshell 120SB-C18柱(100mm×4.6mm,2.7μm)分离,以乙腈-0.1%三氟乙酸水溶液(15∶85,V/V)为流动相,在254nm下检测。该方法可以将鞣花酸与基质良好分离,在0.1~20μg/mL浓度范围内线性良好(r=0.99981);最低检出浓度为0.0134μg/g,回收率为85.3%~95.1%,相对标准偏差小于10%,可用于草莓中游离鞣花酸含量的检测。 An analytical method was developed to extract and determine content of free ellagic acid in strawberry using dispersive solid phase extraction and high performance liquid chromatography (DSPE-HPLC). The free ellagic acid in strawberry was extracted under 0. 1% trifloroacetric acid-methane, and purified by Cl8 dispersive solid-phase extraction. Then strawberry sample was separated by column Poroshell 120 SB-C18 (100 mm×4. 6 mm,2. 7 μm) and measured at UV wavelength of 254 nm with the mobile phase consisting of acetonitrile :water (0. 1% trifloroacetric acid) (15 : 85, V/V). The method could determine the free ellagic acid in strawberry and had fine linear relationship (r=0. 99981) when the range of ellagic acid was within 0. 1-20 μg/mL. Besides that,the minium detection concentration was 0. 0134 μg/g, and average recoveries were 85.3%-95. 1%with relative standard deviation (RSD,%)〈10%. In summary, this approach using DSPE-HPLC was capable to determine free ellagic acid in strawberry.
