黄芪注射液对急性胰腺炎大鼠肠道屏障功能的影响 被引量:3

Effects of astragalus injection on intestinal barrier functions in rats with acute pancreatitis

摘要 目的探讨黄芪注射液对急性胰腺炎大鼠肠道屏障功能的作用及相关机制。方法 50只SD大鼠随机分为假手术组(n=10)、模型组(n=10)、黄芪注射液低剂量组(低剂量组,n=10)、黄芪注射液中剂量组(中剂量组,n=10)、黄芪注射液高剂量组(高剂量组,n=10)。除假手术组外,均采用向胰胆管内输入浓度为5%牛磺胆酸钠溶液方法制备急性胰腺炎大鼠模型,假手术组胰胆管内不注入牛磺胆酸钠,仅对胰腺进行物理翻动和划拨。假手术组和模型组于大鼠尾静脉注射等剂量生理盐水,每6 h 1次,共4次;低剂量组给予黄芪注射液0.5 ml/kg、中剂量组给予黄芪注射液1.0 ml/kg、高剂量组给予黄芪注射液2.0 ml/kg,均为每6 h 1次,共4次。比较各组大鼠小肠黏膜厚度、胰腺组织病理学评分、血清炎症因子和氧自由基水平、B细胞淋巴瘤/白血病-2相关X蛋白(Bax)阳性表达率及组织病理学表现。结果与假手术组比较,模型组小肠黏膜厚度、白介素(IL)-10含量、超氧化物岐化酶(SOD)活性显著下降,胰腺组织病理学评分、IL-6、肿瘤坏死因子-α(TNF-α)和丙二醛(MDA)含量及Bax蛋白表达阳性率显著增加(P均<0.05);低剂量组、中剂量组、高剂量组小肠黏膜厚度、IL-10含量、SOD活性高于模型组,而胰腺组织病理学评分和IL-6、TNF-α、MDA含量及Bax蛋白阳性表达率低于模型组(P均<0.05),且呈剂量依赖性。结论黄芪注射液对急性胰腺炎大鼠肠道屏障功能具有保护作用,其机制可能与降低炎症因子、改善氧自由基及下调Bax蛋白表达相关。 Objective To investigate the effects of astragalus injection on intestinal barrier functions and the related mechanism in rats with acute pancreatitis. Methods Fifty SD rats were randomly divided into five groups (n = 10 each) :sham operation group, model group, low-, medial-, high-dose astragalus injection groups ( low-, medial-, high-dose groups). The rat model of acute pancreatitis was established by input of 5% sodium taurocholate into the pancreaticobiliary duct in all groups except sham operation group performing pancreas mechanical turning only. Astragalus injections of 0.5 ml/kg, 1.0 ml/kg and 2.0 ml/kg were respectively given via tail intravenous injection in low-, medial- and high-dose group, and of normal saline of equal volume were given via tail intravenous injection in sham operation group and model group ( once every 6 h, a total of 4 times for all groups). The intestinal mucosal thickness, the pathological score of pancreas tissues, the serum contents of inflammatory factors and activity of oxygen free radical, the positive expression rate of Bax protein and his- topathologieal findings were compared in all groups. Results Compared with the sham operation group, the intestinal mu- cosa thickness ,serum content of interleukin(IL)-10 and superoxide dismutase(SOD) activity in model group significantly decreased( all P 〈 0.05), and the pathological score of pancreas tissues, the serum contents of IL-6, tumor necrosis factor (TNF)-α and malondialdehyde( MDA), the positive expression rate of Bax protein in model group significantly increased ( all P 〈0.05 ). Compared with model group, the intestinal mucosa thickness, serum content of IL-10 and SOD activity in low-, medial-, high-dose groups significantly increased with dose-dependent manner ( all P 〈 0.05 ) , and the pathological score of pancreas tissues, the serum contents of IL-6,TNF-α and MDA, the positive expression rate of Bax protein in low-, medial-,high-dose groups significantly decreased ( al
