GSTT1和GSTM1基因多态性对煤焦油沥青接触工人多环芳烃代谢的影响

Effects of genetic polymorphisms of GSTT1 and GSTM1 on metabolism of Polycyclic aromatic hydrocarbon in coal tar pitch workers

摘要 目的研究GSTFl和GSTMl基因多态性对煤焦油沥青工人多环芳烃代谢的影响,探讨煤焦油沥青职业接触生物标志。方法选取煤焦油沥青的生产和使用企业中煤焦油沥青职业接触工人374例为接触组、后勤人员135例为内对照组,用液质法测定尿中代谢产物1-羟基芘(1-pyrenol,1-OHP)、1-羟基萘(1-naphthol,1-OHN)、2.羟基萘(2-naphthol,2-OHN)的浓度,采用多重PCR技术检测全血中GSTT1和GSTM1的基因多态性。结果吸烟组尿中1-OHN、1-OHP、2-OHN含量高于不吸烟组,接触组尿中1.OHN、1-OHP、2-OHN含量高于对照组,差异均有统计学意义(P〈0.05)。携带GSTM1和GSTTI基因型工人尿中1-OHN、1-OHP、2-OHN含量高于携带阴性基因型者,差异有统计学意义(P〈0.05)。不同工种对象尿中1-OHP含量的差异有统计学意义(P〈0.05),班后尿中1-OHP、1-OHN、2-OHN含量高于班前,差异有统计学差异(P〈0.05)。携带GSTT1基因型和GSTM1基因型与尿中代谢产物的含量有正相关关系(P〈0.05)。结论携带GSTF1基因型和GSTM1基因型有利于多环芳烃在体内的代谢。 Objective To investigate the influence of genetic polymorphisms of GSTR1 and GSTM1 on metabolism of polycyclic aromatic hydrocarbon (PAHs) in coal tar pitch workers and to explore the effeetive biomarker of occupational exposure to coal tar pitch. Methods 374 cases of workers exposed to coal tar pitch work at the enterprise which use and product coal tar pitch in Shan Dong province named as exposure group.The administrative and the support staff 135 cases were selected as control group.The concentration of the metabolic product in urine by HPLC/MC.The genetic polymolphisms of GSTT1 and GSTM1 were detected by multiple PCR method. Results The content of metabolic product in smoke group were significantly higher than workers do not smoke (P〈0.05). The content of metabolic product in exposure group were significantly higher than control group (P〈0.05). The content of metabolic product in workers carry positive GSTT1 and GSTM1 were significantly higher than workers carry negative one (P〈0.05). The content of metabolic product in different types of labor were significantly different (P〈0.05). The content of metabolic product were significantly higher in urine after work than before work (P〈0.05). The content of metabolic product and workers can'y positive GSTT1 and GSTM1 had a positive correlation (P〈0.05). Conclusion Carrying positive GSTT1 and GSTM1 promote polycyclic aromatic hydrocarbons in the body' s metabolism.
