摘要
目的观察肾病Ⅰ号方总皂苷对单侧输尿管结扎(UUO)大鼠肾纤维化的影响,阐明其抗肾纤维化的作用机制,探索中药复方药效物质基础的研究思路。方法建立UUO大鼠模型,30只大鼠随机分为5组,每组6只:正常组、模型组、贝那普利组、总皂苷组、肾病Ⅰ号方组。于术后两周随机处死大鼠,取左侧肾组织。观察干预后各组大鼠一般情况变化,测定24小时尿蛋白定量、血清肌酐(Scr)、血清尿素氮(BUN)的含量,光镜下观察肾组织纤维化的病理改变,免疫组织化学法检测肾组织胶原Ⅳ(COL-IV)的表达情况。结果肾病Ⅰ号方总皂苷能显著降低Scr、BUN,减轻尿蛋白,减少细胞外基质COL-IV的阳性表达(P<0.05),改善肾小球硬化程度,与贝那普利组和肾病Ⅰ号方组比较无明显差异(P>0.05)。结论肾病Ⅰ号方总皂苷可以抑制细胞外基质COL-IV的合成,减少细胞外基质的积聚,从而达到抗肾纤维化的作用。
Objective To observe the effect of "The Kidney Disease I Saponins"on renal fibrosis by unilateral ureteral( UUO) in rats. Which aims to illustrate its resistance of renal fibrosis mechanism and explore the research tracks about material foundation of efficacy of Chineae herbs compound prescription. Methods Establish a rat model of UUO,30 rats were divide into 5 groups named normal control,model control,benazepril control,saponins control,kidney disease I control. Rats of every experimental group was sacrified randomly at two weeks postoperative and taken the left renal tissue. Observing the general changes in rats after the intervention,measure the 24- hour urinary protein quantitative,the content of serum creatine( Scr),blood urea nitrogen( BUN),the pathological changes of renal tissue fibrosis was observe under ligh microscope,the expression of renal tissue collagen IV( COL- IV) were detected by immunohistochemical staining. Results "The kidney Disease I saponins"can help Scr,BUN significant reductions,decrease urinary protein,reduce the positive rates of extracellular matrix( P〈0. 05). To improve the change of glomerular sclerosis. Therewas no significant difference between benazepril control and kidney disease I control( P〉0. 05). Conclusion The kidney disease I saponins can inhibit the synthesis of extracellular matrix COL- IV and to reduce the accumulation of extracellular matrix so as to achieve the role of resistance of renal fibrosis.
