柿叶黄酮对阿霉素肾病鼠肾组织纤维连接蛋白表达的影响

摘要 目的探讨柿叶黄酮(PLF)对阿霉素肾病大鼠肾组织纤维连接蛋白(FN)表达及对肾脏免疫病理损伤的影响。方法制备阿霉素肾病鼠模型,采用PLF干预模型鼠,测定模型鼠各阶段24 h尿蛋白变化(于模型制备前、制备后1周、给药3周及给药5周后测定),应用苏木精-伊红(HE)染色法观测肾脏病理形态变化,用免疫组化法观测肾组织中FN表达。结果 PLF能明显降低模型鼠24 h尿蛋白水平(P<0.01)、改善肾脏病理损伤以及抑制肾组织FN表达(P<0.01)等。结论 PLF能通过降低模型鼠肾组织FN表达、改善肾组织纤维化,对模型鼠肾脏起到一定的保护作用。 Objective To investigate the effect of persimmon leafflavonid(PLF) on the expression offibronectin(FN) in kidney of adriamyein nephropathy(AN) rat. Methods AN rat model was prepared. 24 h urine protein(24HUP) was detected in different time points (before ADR injection, 1 week afterinjection ,3,5 weeks after treatment). The pathomorphological changes of kidney were observed by HE staining. The expression of FN was detected by immunohistochemieal method. Results PLF could obviously reduce 24HUP,improve the pathologic damage of kidneys and inhibit the expression of FN in kidney of AN rats. Con- clusion PLF can play certain protective role on the kidney of AN rat by inhibiting tissue fibrosis and down-regulating expression of FN.
