摘要
我们应用细胞培养及免疫组化的方法,探讨了肾小球系膜细胞增生及系膜硬化的机制,并进而观察了它们在肾小球硬化中的作用。结果显示:系膜细胞有自分泌功能,肿瘤坏死因子和血管内皮素可促进系膜细胞增生。大量增多的系膜基质不但含有Ⅳ型胶原,也有Ⅲ型胶原。系膜细胞增生、系膜基质增多最终导致肾小球硬化。
he role of glomerular mesangial cells(MC)and mesangial matrix(MM)in glomerular sclerosis was studied by using rat mesangial cell cuIture, immunohistochemical studies for laminin, laminin receptors as well as collagen types Ⅳ and Ⅲ.The effect of tumor necrosis factor(TNF) and endothelin(ET)on MC was also studied. Results show that TN F and ET have potent mitogenic effects on MC.The glomerular MC possess the special capability of secreting TNF and ET. TNF and ET can also in- fluence and induce the secretion of each other. The MM is increased in the MC nodular colonies formed after prolonged culture. These colonies are composed of collagen Ⅳ and collagen Ⅲ.Therefore,in-juring factors can cause MC and MM to proliferate and produce glomerulosclerosis.
