摘要
目的:观察肾病Ⅰ号方(SBYHF)对体外培养的大鼠肾小球系膜细胞(glomerular mesangial cell,GMC)的增殖及表达IV型胶原(COL-Ⅳ)及结缔组织生长因子(CTGF)的影响,探讨其抗肾纤维化的机制。方法:通过体外培养GMC技术及体外药物血清实验方法,采用CCK-8法检测GMC的增殖,ELISA法检测COL-Ⅳ的分泌水平,RT-PCR法检测CTGF mRNA的表达。结果:肾病Ⅰ号方可显著抑制GMC的增殖(P<0.01),减少COL-Ⅳ的分泌和CTGF mRNA的表达(P<0.01)。结论:肾病Ⅰ号方可能通过减少GMC的COL-Ⅳ的分泌,下调CTGF mRNA表达,而抑制GMC增殖,从而发挥抗肾脏纤维化的作用。
Objective: To explore the molecular and cellular mechanism of SBYHF in ameliorating renal fibrosis through observing the effect of proliferation andexpression ofCOL-IV and CTGFin Rat Glomerular Mesangial Cells.Methods: The proliferation of Rat GMC fostered in SBYHF was detected by CCK-8, the COL-IV secretion was detected by ELISA, and the CTGF was detected by Real Time PCR. Results: SBYHF significantly inhibited GMC proliferation(P〈0.01) and decreased the expression of COL-IV and CTGF (P〈0.01). Conclusion: SBYHFmay through reduces the COL-IV secretion and down-regulated the expression ofCTGF to inhibit the proliferation of Rat Glomerular Mesangial Cells. This may be the mechanism of SBYHF in ameliorating renal fibrosis.
