摘要
目的探讨联合应用IL-4突变体及抗IL-17抗体对支气管哮喘(简称哮喘)小鼠炎性细胞及细胞因子水平的影响。方法40只BALB/c雌性小鼠随机分为对照组、哮喘组、IL-4突变体治疗组、抗IL-17抗体治疗组、联合IL-4突变体及抗IL-17抗体治疗组(简称联合治疗组)。哮喘组和治疗组用鸡卵白蛋白(ovalbumin,OVA)和氢氧化铝致敏,用OVA激发,建立小鼠哮喘模型,对照组用生理盐水代替,其中IL-4突变体治疗组、抗IL-17抗体治疗组及联合治疗组分别于诱发前30min给予腹腔注射相关抗体蛋白,对照组及哮喘组用生理盐水代替。细胞计数板测外周血炎性血细胞,酶联免疫吸附试验双抗体夹心法测定各组血清细胞因子水平变化。结果与对照组比较,哮喘组炎性细胞数及IL-4、IL-17、IgE水平明显升高(P〈0.05);联合治疗组可有效降低炎性细胞计数及IL-4、IL-17、IgE水平,与哮喘组比较差异有统计学意义(P〈0.05);与单独治疗组相比,联合治疗组效果更明显(P〈O.05)。结论联合应用IL-4突变体及抗IL-17抗体对哮喘小鼠外周血炎性细胞有抑制作用,且可以降低IL-4、IL-17、IgE细胞因子水平,为治疗和缓解哮喘提供-定的帮助。
Objective To observe the effects of IL-4 mutant combined with anti IL-17 antibody on inflammatory cells and cytokines level of asthmatic mice. Methods Forty female BALB/c mice were randomly divided into 5 groups:the control group, the asthma group, the IL-4 mutant treatment group, the anti IL-17 antibody treatment group, the combination of IL-4 mutant and anti IL-17 antibody treatment group. Mice in asthma group and treatment group were sensitized and challenged by OVA and AL(OH)3, and challenged with OVA to establish asthmatic models, while mice in the normal group were treated with saline. Mice in the IL-4 mutant treatment group, the anti IL-17 antibody treatment group and the combined treatment group were injected intraperitoneally with the related antibody 30 min before challenged, while mice in normal group and asthmatic group received saline. Then peripheral blood cell count plate test inflammatory cells, and the cytokines level of serum were measured by ELISA. Results Compared with control group, inflammatory cells and IL-4, IL-17, IgE in the asthma group were significantly increased ( P 〈0.05); Combined treatment group can effectively reduce the inflammatory cell count and IL-4, IL 17, IgE level, compared with asthma group difference was statistically significant (P 〈0. 05) ;Compared with the separate treatment group, the combined treatment group effect is more obvious ( P 〈0.05). Conclusions Combined use of IL-4 mutant and anti IL-17 antibody for bronchial asthma mice showed inhibitory effect on the peripheral blood of inflammatory cells, and can reduce the cytokine levels of IL-4, IL-17, IgE, provide certain help for the treatment and ease the hronchial asthma,
