摘要
[目的]研究牛奶中甲氧苄啶的胶体金快速检测装置,快速筛查牛奶中甲氧苄啶添加物含量并评价牛奶品质。[方法]制备抗原,免疫得到抗体并摸索胶体金免疫层析装置的样品检测性能。[结果]试验所得装置对牛奶中甲氧苄啶的肉眼可见检测线为2 ng/L,假阳性率为0%,假阴性率为0%;与常见3种混合添加物无交叉反应,特异性高;不同批次间结果重复性高度一致;稳定性好,易于保存,在37℃条件可存放35 d,50℃存放6 d对其质量无影响;兼容性好,适合6种不同种类奶基质;检测时间短,仅需6 min。[结论]该研究的装置检测时间短,使用简单,重复性好,产品稳定性高,适用于牛奶中甲氧苄啶的现场快速筛查和检测。
[Objective] To research a rapid detection system for trimethoprim in milk,and to provide methods for the rapid screening of milk quality and the trimethoprim content in milk. [Method] Antigen was prepared. Antibody was obtained through immunity. Sample detection performance of colloidal gold immunochromatography system was found out. [Result] This system had a detection limit as low as 2 ng / L,and the false positive rate and false negative rate were both 0%. There was no cross reaction with common three mixed additives with high specificity. Result of repeatability in different batches was highly consistent. This system had high stability and was easy to preserve. The quality was not affected when milk was stored at 37 ℃ for 35 d or 50 ℃ for 6 d. [Conclusion]This system has short detection time,is simple,repeatable and stable,and is suitable for the rapid screening and detection of trimethoprim in milk.
