摘要
以8种不同基因型的萝卜为试验材料进行带柄子叶离体培养,筛选诱导不定芽和不定根的最佳培养基配方。结果表明:喜诺晓青和翠绿萝卜具有高频再生潜力,由其带柄子叶诱导不定芽的最佳培养基为:MS培养基+30 g·L-1蔗糖+0.7%琼脂+5 mg·L-16-BA+0.05 mg·L-1NAA,诱导不定根的最佳培养基为:MS培养基+30 g·L-1蔗糖+0.7%琼脂+5 mg·L-16-BA+0.10 mg·L-1NAA。
To select the best hormone combination for inducing adventitious shoots and adventitious roots,the in vitro culture of cotyledons with petiole from 8 genotypes of radish were carried out in this research. The results showed that Xinuoxiaoqing and Cuilü radish had high- frenquency regeneration potential. The optimum medium for their adventitious shoot induction was MS + 30 g·L- 1Sucrose + 0. 7% Agar + 5 mg·L- 16- BA + 0. 05 mg·L- 1NAA,and that for adventitious root induction was MS + 30 g·L- 1Sucrose + 0. 7% Agar + 5 mg·L- 16- BA +0. 10 mg·L- 1NAA.
