摘要
为了获得番茄离体再生体系的优化方案,对离体再生体系中番茄无菌苗的获得、生芽培养基激素组合及诱导出芽、生根培养基激素组合及诱导生根进行了一系列优化试验.结果显示,在浸泡、暗培养条件下,无菌苗生长快速、整齐;生芽培养基以 MS+2.0 mg/L BA+0.02 mg/L IAA 为宜;在再生芽个数方面,子叶要优于真叶、下胚轴,但由于下胚轴具有体积小、操作方便、形成愈伤组织和根的速率较快、根系发达等优点,因此外植体可选下胚轴.下胚轴生根培养基以 MS+0.3 mg/L IAA 为宜,子叶、真叶则使用 MS+0.1 mg/L BA+0.3 mg/L IAA 为宜.再生苗在无菌处理的移栽土中成活率较高,可达 90%.
A series of experiments are made on how to attaining tomato axenic seedlings,filtrate hormone constitution on shoot and root induce and induce shoot and root in order to obtain the best way on regenerative system of tomato .The results show that tomato axenic seedlings grow rapidly and tidily after seeds are soaked and trained in dark.MS+2.0 mg/L BA +0.02 mg/L IAA is the best shooting medium.More regenerative buds are obtained on the calli of cotyledons than on those of euphyllas and hypocotyls,but because hypocotyls are easily obtained and grow faster in callus tissuse and shooting,hypocotyls are elected as the explant.MS+0.4 mg/L BA is considered as the best hormone constitution on root induction of hypocotyls.However,a small quantity of IAA(0.1 mg/L) is added to make cotyledons grow better.Transplanted plants can survive when soil is sterilized,with the survival rate of 90% in the sterilized soil higher than that in the unsterilized soil.
