正交试验法优化长春胶囊的提取工艺 被引量:1

摘要 目的采用多重指标,优选长春胶囊的提取工艺,为其工业化生产提供依据。方法:采用紫外-可见分光光度法分别测定总皂苷、总黄酮及总多糖的含量。以总皂苷、总黄酮含量的综合评分为评价指标,采用L_9(3~4)正交试验法考察乙醇浓度、加醇倍量、提取时间对脂溶性成分提取工艺的影响;以总多糖含量为评价指标,采用L_9(3~4)正交试验法考察加水倍量、提取时间、提取次数对水溶性成分提取工艺的影响,优化长春胶囊的提取工艺。结果脂溶性成分提取的最佳工艺为10倍量65%乙醇回流提取三次,每次1.5h;水溶性成分提取的最佳工艺为8倍生药量水煎煮二次,每次1.5h。结论验证试验表明,该提取工艺稳定可行、重复性良好。研究为长春胶囊的工业化生产提供了依据。 AIM Using multiple indicators, optimizing extraction technology of changchun capsule and to provide basis for its industrialized production. METHODS: UV-visible spectrophotometry determination of total saponins, total flavonoids and total polysaccharides of. In total saponins, total flavonoids in the composite score for the index, using L_9(3~4) orthogonal experiment investigated ethanol concentration, the amount of ethanol, extraction time extraction process of fat-soluble ingredients; in total polysaccharide content evaluation, using L_9(3~4) orthogonal experiment study times the amount of water, extraction time, extraction times extraction of water-soluble components influence, optimization of extraction process Changchun capsules. RESULTS The optimum extraction of fat-soluble ingredients was 10 times the amount of 65% ethanol extracted three times,each time 1.5h;the optimum extraction of water-soluble components is 8 times the amount of crude drug boiling water twice, each time 1.5h.CONCLUSION: The experiment indicated that the extraction process is stable, good repeatability. Changchun capsule study provided the basis for industrial production.
