葡聚糖凝胶色谱结合高速逆流色谱提取蓝莓中花青素 被引量:4

Separation and Preparation of Anthocyanins from Blueberry by High Speed Count Current Chromatography(HSCCC) and Sephadex Chromatography

摘要 试验结合葡聚糖凝胶色谱及高速逆流色谱对野生蓝莓中的花青素进行了分离纯化。蓝莓粗提物先经过葡聚糖凝胶色谱初步分离,得到花青素含量高的组份,再经高速逆流色谱分离,以MTBE-正丁醇-乙腈-水(体积比1︰3︰1︰5)为两相溶剂体系,流速0.5 m L/min、主机转速1 860 r/min以及检测波长280 nm的条件下进行分离,从蓝莓的葡聚糖凝胶色谱柱分离产物中一次性分离得到两种花青素,经超高效液相色谱分析,2种化合物的纯度分别为65.0%和90.0%。 Two anthocyanins were isolated from blueberry by the combination of sephadex chromatogram and high speed count-current chromatography (HSCCC). The crude extract of blueberry was separated by sephadex chromatogram, and the fraction containing anthocyanins was obtained. Then the fraction was separated by HSCCC with MTBE-n-butyl alcohol- acetonitrile-water (volume ratio 1 : 3 : 1 : 5) as the two-phase solvent systems. Under the conditions of flow rate of 0.5 mL/ min, while the equipment rotated at 1 860 r/min and the detection wavelength was 280 um. Two kinds of anthocyanins were obtained from the fraction of blueberry by sephadex chromatogram in one-time. The purities of the two compounds determined by UPLC were 65.0% and 90.0%, respectively.
