大黄苷元抗大鼠脑缺血损伤及对炎性细胞因子的影响 被引量:18

Effects of rhubarb aglycone antagonising cerebral ischemic injury and influence on inflammatory factors in rats with cerebral ischemia

摘要 目的:探讨大黄苷元抑制脑缺血炎性级联反应的机制。方法:60只大鼠随机分为假手术组、模型组、尼莫地平组及大黄苷元低、中、高剂量组。用线栓法制备大鼠局灶性脑缺血模型;观察神经症状积分、脑组织含水量和梗死面积;用放射免疫法测定脑组织肿瘤坏死因子α(TNFα)、白细胞介素1(βIL 1β)、转化生长因子β(TGFβ)水平;用免疫组化法测定TNFα、血管细胞黏附因子1(VCAM 1)表达;用原位杂交法测定VCAM 1 mRNA表达。结果:与假手术组比较,模型组大鼠神经症状积分、脑含水量及梗死面积均增加;脑组织TNFα和IL 1β水平增高,TGFβ水平降低,TNFα和VCAM 1表达增强(P均<0.01)。与模型组比较,尼莫地平组和大黄苷元各剂量组神经症状积分、脑含水量及梗死面积均明显降低,大黄苷元低剂量组神经症状积分降低尤为显著;大黄苷元低剂量组TNFα和IL 1β水平及TNFα、VCAM 1表达均明显降低,TGFβ水平增高;尼莫地平组TNFα和IL 1β水平及VCAM 1表达均明显降低,TGFβ水平显著增高(P均<0.01),大黄苷元低剂量组TNFα、IL 1β水平及TNF表达降低和TGFβ水平增高均较尼莫地平组显著。结论:由TNFα、IL 1β、VCAM 1介导的炎性级联反应增强及TGFβ保护作用减弱是脑缺血损伤的重要机制。大黄苷元抗脑缺血损伤的作用机制可能与其抑制炎性级联反应和提高脑保护因子水平有关。 Objective:To investigate the mechanism of rhubarb aglycone on inhibiting inflammatory cascade reaction in cerebral ischemic injury rats. Methods: Sixty rats were randemly divided into sham - operated group, model group, nimodipine group and low, middle, high dosage of rhubarb aglycone groups. Focal cerebral ischemic model was replicated by cerebral middle artery occlusion method with nylon thread. The changes of score of neural symptoms, brain water content and cerebral infarction area were observed after 6 - hour ischemia, meanwhile the levels of tumor necrosis factor - α(TNF - α), interleukin -1β(IL - 1β), transforming growth factor - 1β(TGF - 1β) of rats brain tissues, the expressions of TNF - α and VCAM - 1, the expression of vascular cellular adhesion molecule (VCAM - 1) mRNA were measured. Results: Compared with sham -operated group, the score of neural symptoms, brain water content and cerebral infarction area of rats in model group as well as the levels of TNF -α and IL - 11β of rats brain tissue in model group increased, and the level of TGF -1β decreased, while the expressions of TNF -α and VCAM - 1 increased(all P〈0.01). Compared with the model group, the score of neural symptoms, brain water content and cerebral infarction area of rats in every treatment group decreased significantly, especially the score of neural symptoms decreased more obviously in rhubarb aglycone low dosage group. The levels of TNF -α, 1L - 1β and the expression of TNF - α , VCAM - 1 in rats' brain tissue of rhubarb aglycone low dosage group decreased, while the expression of TGF - 1β increased. The levels of TNF -α, IL - 1β and the expression of VCAM - 1 in rats brain tissue in nimodipine group decreased, while the level of TGF - 1β increased. Compared with nimodipine group, the decrease of the levels of TNF -α, IL - 11β, the expression of TNF -α as well as the increase of the TGF - 1β level of rats’ brain tissue in low dosage rhubarb aglycone group were more significant (a
