摘要
目的:研究β-榄香烯对紫杉醇耐药肺腺癌细胞A549/Taxol的抑制作用及其对Wnt/β-catenin信号转导通路的影响。方法:采用浓度梯度诱导法制备紫杉醇耐药肺腺癌细胞株A549/Taxol。以不同浓度β-榄香烯作用于A549/Taxol细胞48 h,采用CCK8法检测细胞增殖;实时定量PCR法检测多药耐药基因(MDR1)及β-catenin、Survivin基因的mRNA表达;Western blot法检测A549/Taxol细胞P糖蛋白(P-gp)及β-catenin、Survivin的蛋白表达。结果:与对照组比较,β-榄香烯呈浓度相关性抑制A549/Taxol细胞增殖;β-榄香烯(20、40、80μg/ml)作用48 h后,A549/Taxol细胞MDR1、β-catenin、Survivin mRNA及蛋白表达水平均明显下降(P<0.01)。结论:β-榄香烯可能通过下调β-catenin、Survivin mRNA及其蛋白的表达,影响Wnt/β-catenin信号转导通路,发挥抑制A549/Taxol细胞增殖的作用;β-榄香烯对肺腺癌细胞紫杉醇耐药性具有拮抗作用,该作用可能通过抑制Wnt/β-catenin信号转导通路实现。
Objective: To study the effect of β-elemene on inhibiting function and Wnt / β-catenin signaling pathway of paclitaxel-resistant human lung caner cell A549 / Taxol. Methods: Paclitaxel-resistant human lung cancer cell line( A549 /Taxol) was established with A549 cells. A549 / Taxol cells were treated with different concentrations of β-elemene. The cellular proliferation was detected by CCK-8 assay. The expression of MDR1,β-catenin,Survivin mRNA in A549 / Taxol cells were measured by QT-PCR. The expressions of P-gp,β-catenin,Survivin protein were detected by Western blot. Results:Compared with control group, β-catenin could inhibit the proliferation of A549 / Taxol cells according to different concentrations. After treatment with 20,40,80 μg / m L ofβ-elemene for 48 h,the expression levels of MDR1,β-catenin,Survivin mRNA and protein were decreased significantly( P〈0. 01). Conclusion: β-elemene could inhibit the proliferation of A549 / Taxol cells,associating with down-regulation expression of β-catenin,Survivin mRNA and their proteins,which may be related with the inhibition of Wnt / β-catenin signaling pathway.
