参芪扶正注射液对人肺腺癌耐药细胞株A549/DDP的增敏作用 被引量:10

The Sensitization Effect of ShenQiFuZheng Injection on Human Lung Adenocarcinoma Cancer Cell Line A549 / DDP

摘要 目的:观察参芪扶正注射液(参芪液)对人肺腺癌顺铂耐药细胞株(A549/DDP)耐药性的逆转作用及其可能机制。方法:1~120 g/L参芪液干预A549/DDP细胞48 h,倒置显微镜观察细胞的形态变化;MTT法检测参芪液对A549/DDP细胞株增殖的作用以及对顺铂(DDP)敏感性的影响;低浓度参芪液(8 g/L)、中浓度参芪液(16 g/L)、高浓度参芪液(32 g/L)联合DDP(40μg/ml)分别作用于A549/DDP细胞24 h,流式PI单染法检测细胞凋亡率,Western blot检测Cleaved-caspase-3蛋白表达。结果:与对照组比较,2~120 g/L参芪液对A549/DDP细胞有增殖抑制作用,且呈浓度依赖性(P〈0.05);2 g/L参芪扶正注射液作用A549/DDP细胞后,顺铂敏感性提高,逆转倍数(RF)为2.66倍;与DDP组比较,低、中、高浓度参芪液联合DDP组,均能显著诱导A549/DDP细胞凋亡(P〈0.05),Cleaved-caspase-3蛋白表达上调。结论:参芪扶正注射液能够逆转A549/DDP细胞对顺铂的耐药性,其机制可能是通过启动凋亡调控基因介导的Caspase级联反应,活化Caspase-3,从而导致细胞凋亡。 Objective: To investigate the reverse effect and mechanisms of Shen Qi Fu Zheng injection( SQFZI) on the cisplatin resistance of human lung adenocarcinoma cancer cell line( A549 / DDP). Methods: 48 h after treatment A549 / DDP cell with 1-120 g / L SQFZI,inverted microscope was used to observe the morphological changes of A549 / DDP cell line; MTT assay was used to determine the effect of SQFZI on proliferation and sensitiveness to cisplatin of A549 / DDP cell. 24 h after treatment A549 / DDP with low-( 8 g / L),middle-( 16 g / L),high-( 32 g / L) concentration of SQFZI combined with DDP( 40μg /m L),PI Flow cytometry test and Western blot were used to determine cell apoptosis and Cleaved-caspase-3 expression respectively. Results: Compared with control group,2 ~ 120 g / L SQFZI can inhibit the proliferation of A549 / DDP cell in a concentration-dependent manner( P 〈 0. 05). Treated with 2 mg / m L SQFZI,the sensitivity of A549 / DDP cell line to cisplatin increased and the reversal fold up to2. 66. Compared with DDP group,SQFZI at different concentration combined with DDP significantly induced apoptosis in A549 / DDP cells( P 〈 0. 05) and the expression of Cleaved-caspase-3 was up-regulated.Conclusion: SQFZI could moderately reverse the drug resistance of A549 / DDP cells to cisplatin. The mechanism may be related to start-up Caspase cascade reaction mediated by apoptosis regulation gene and activate Caspase-3,which leads to apoptosis.
